摘  要:目的:选用宁夏枸杞不同部位为外植体,探究不同外植体、不同灭菌时间、激素配比不同对于宁夏枸杞组织培养的影响,测定其蛋白质含量并与野生种对比。方法:以枸杞的嫩叶、嫩茎为外植体,探讨培养基类型、植物激素类型、激素浓度、消毒时间、接种方式、等对培养结果的影响,为优化枸杞快速繁殖体系提供依据,并通过测定紫外分光光度计对野生枸杞和栽培枸杞蛋白质含量进行对比和分析。结果:最佳消毒试剂及时间:75%酒精30 s+0.1%升汞8 min;以MS培养基为基本培养基,诱导丛生芽分化的最佳培养基:NAA0.5mg/L+6-BA0.5mg/L,诱导率为86.67%,死亡率为13.33%;诱导芽增殖的最佳培养基:NAA0.5mg/L+6-BA0.5mg/L,诱导率为76.67%,死亡率为23.33%;最佳生根培养基:NAA0.6mg/L+6-BA1.5mg/L,大量生根且根系浓密,栽培枸杞比野生枸杞的蛋白质含量明显增多。Objective: Using the different parts of Lycium barbarum from Ningxia as explants,discussing the effects of different explants,different disinfection time and hormone concentration on process of tissue culture,determined the change of protein content. Methods: Using the young leaves and tender stems as explants,the effects of media type,plant hormone type,hormone concentration,disinfection time,inoculation method,etc. on culture results were explored to provide basis for optimizing the rapid propagation system of Lycium barbarum,and by measuring the absorbance to compare and analyzed the protein in wild and cultivated barbarum. Result: The Best disinfection reagents and time: 75% alcohol 30 s + 0. 1% Hg Cl28 min; MS medium as basic medium,the best medium for inducing bud differentiation: NAA0. 5 mg/L + 6-BA0. 5 mg/L,the induction rate is 86. 67% and the mortality rate is 13. 33%; the optimal medium for inducing shoot proliferation: NAA0. 5 mg/L + 6-BA0. 5 mg/L,the induction rate is 76. 67% and the mortality rate is 23. 33%; the best rooting medium: NAA0. 6 mg/L + 6-BA1. 5 mg/L,multi-root and roots thick,the protein content of cultivated Lycium barbarum is significantly higher than wild earthworms.
