摘 要:目的:针对钛种植体植入后组织感染问题,在纯钛阳极氧化-植酸处理工艺基础上,引入具有抗菌性的铜元素,探究不同镀铜含量涂层的细胞相容性以及对抗菌性能的影响。方法:纯钛表面两步法阳极氧化、植酸及化学镀铜制备涂层,通过SEM及元素分析,激光共聚焦显微镜和CCK-8实验检测各组涂层成骨细胞的细胞活性,采用吖啶橙/溴化乙锭(AO/EB)双重荧光染色法检测抗菌性。结果:随着镀铜时间的增加铜含量增多,涂层微孔孔隙变小变少,涂层表面愈加致密。各组均有利于成骨细胞生长,无细胞毒性,载铜后各组试件表面细胞伸展均较好,其中镀铜时间6min的Cu3组细胞整体伸展最好;载铜后涂层具有抗菌性,随着镀铜含量的增加,其中Cu3组抗菌率达86%,明显优于其他组。结论:纯钛阳极氧化-植酸-低载铜复合膜层具有良好的细胞相容性,抗菌能力随着铜含量的增多而增强,涂层表面含铜量为1.2wt%,具有较强抗菌能力。Objective:Aiming at tissue infection after titanium implants implantation,on the basis of pure titanium anodization phytic acid treatment process,the copper elements with antibacterial properties were introduced to explore the cytocompatibility and antibacterial properties of different copper plated coatings.Methods:Coating on pure titanium surface by two step anodizing,phytic acid and electroless copper plating.Through SEM and element analysis;Detection of cell activity of osteoblasts in each group by laser confocal microscopy and CCK-8 test.Acridine orange/ethidium bromide(AO/EB)double fluorescent staining was used to detect the antibacterial.Results:With the increase of copper plating time,the pore size of the coating becomes smaller and smaller,and the surface of the coating is more compact.All groups were beneficial to the growth and cytotoxicity of osteoblasts.After loading copper,the surface cells of each group were well spread,and the Cu3 group with copper plating time of 6 min had the best overall extension.The coating has antibacterial properties after copper loading,with the increase of copper content.Among them,the antibacterial rate of group Cu3 was 86%,which was obviously superior to other groups.Conclusion:Pure titanium Anodizing-phytic acid low load copper composite film has good cytocompatibility.The antibacterial ability is enhanced with the increase of copper content.When the copper content on the surface of the coating is 1.2 wt%,it has strong antibacterial ability.
