头痛宁胶囊对偏头痛大鼠血清多巴胺、多巴胺-β-羟化酶、去甲肾上腺素水平的影响  

The Influence of Toutongning Capsule on the Levels of Serum Dopamine,Dopamine-β-Hydroxylase and Norepinephrine in Rats with Migraine

摘　　要：目的观察头痛宁胶囊对偏头痛大鼠血清多巴胺(DA)、多巴胺-β-羟化酶(Dβ-H)、去甲肾上腺素(NE)水平的影响。方法将36只健康雄性Wistar大鼠普通喂养1周,随机分为空白对照组(6只)和偏头痛模型组(30只),空白对照组给予生理盐水皮下注射,偏头痛模型组皮下注射硝酸甘油(NTG)制备大鼠偏头痛模型,并随机分为模型对照组、预防高剂量组、预防低剂量组、治疗高剂量组、治疗低剂量组,每组6只。结果预防高剂量组60min及180min血清Dβ-H含量与空白对照组比较,差异无统计学意义(P>0.05),预防高剂量组60min血清NE含量与空白对照组比较,差异无统计学意义(P>0.05);其余各组两两比较,差异有统计学意义(P<0.05)。DA与NE存在负相关性,DA与Dβ-H存在负相关性,NE与Dβ-H存在正相关性。结论头痛宁胶囊可能通过增加Dβ-H活性,降低DA水平,升高NE水平,发挥治疗及预防偏头痛的作用,且其对偏头痛的预防作用大于治疗作用。Objective To observe the effect of Toutongning capsule（TTNC）on the levels of serum dopamine（DA）,dopamine-β-hydroxylase（Dβ-H）,and norepinephrine（NE）in rats with migraine.Methods Thirty-six healthy male wistar rats were fed normally for 1 week,and randomly divided into blank control group（n=6）and migraine model group（n=30）.Rats in the blank control group were given subcutaneous injections of normal saline.Rats in the migraine model group were treated with subcutaneous nitroglycerin（NTG）to establish the migraine models,and randomly divided into model control group（n=6）,high-dose TTNC prevention group（n=6）,low-dose TTNC prevention group（n =6）,high-dose TTNC treatment group（n =6）,and low-dose TTNC treatment group（n=6）.Results There was no significant difference in serum Dβ-H between the high-dose TTNC prevention group and the blank control group after 60 min and 180 min of the last administration（P〈0.05）.There was no significant difference in serum NE content between the high-dose TTNC prevention group and the blank control group after 60 min of the last administration（P〈0.05）.Comparing with other groups,the difference was statistically significant between two groups（P〈0.05）.There was a negative correlation between DA and NE,a negative correlation between DA and Dβ-H,and a positive correlation between NE and Dβ-H.Conclusion TTNC may play a role in the treatment and prevention of migraine by increasing the activity of Dβ-H,decreasing the level of DA and increasing the level of NE,and its preventive effect on migraine is better than the therapeutic effect.
