沙格雷酯下调肺动脉高压大鼠右室TRPC1、TRPC6通道及calcineurinA/NFATc3表达

摘　　要：目的研究5-HT_(2A)受体拮抗剂沙格雷酯对肺动脉高压大鼠的右室TRPC1,TRPC6通道及calcineurin A/NFATc3表达的影响。方法 18只大鼠分为对照组、肺动脉高压组和沙格雷酯组,每组6只。对照组给予生理盐水腹腔注射和生理盐水灌胃,肺动脉高压组给予野百合碱腹腔注射及生理盐水灌胃,沙格雷酯组给予野百合碱腹腔注射及沙格雷酯灌胃。干预4周时采用RT-PCR和Western blot检测右室TRPC1、TRPC6通道和calcineurin A/NFATc3的mRNA和蛋白表达,并对三组间结果进行比较。结果与对照组相比,肺动脉高压组大鼠右室心肌中TRPC1、TRPC6通道、calcineurin A/NFATc3的mRNA和蛋白表达升高(P<0.05);与肺动脉高压组相比,沙格雷酯组大鼠右室心肌TRPC6通道、calcineurin A/NFATc3的mRNA表达降低,TRPC1、TRPC6通道、calcineurin A/NFATc3的蛋白表达降低(P<0.05)。结论肺动脉高压大鼠右室心肌的TRPC1、TRPC6通道、calcineurin A/NFATc3表达上调,沙格雷酯可逆转这些蛋白和基因的表达水平。Objective To explore the effect of 5-HT（2A） receptor antagonist sarpogrelate on the expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in the right ventricle of rat with pulmonary hypertension. Methods Rats were divided into control group,pulmonary hypertension group and sarpogrelate group. Rat model of pulmonary hypertension induced by monocrotaline was established,and the sarpogrelate was given intragastrically. The expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in the right ventricle was detected by RT-PCR and Western blot at 4 week. And the results were compared between three groups. Results Compared with control group,the mRNA and protein expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in the right ventricle increased in pulmonary hypertention group（ P〈0. 05）. Compared with pulmonary hypertension group,the mRNA expression of TRPC6 channel and calcineurin A/NFATc3 in the right ventricle decreased in sarpogrelate group,and the protein expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in the right ventricle also decreased（ P〈0. 05）. Conclusion The expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in the right ventricle of rats with pulmonary hypertension is upregulated. Sarpogrelate may downregulate the expression of TRPC1,TRPC6 channel and calcineurin A/NFATc3 in right ventricle of rats with pulmonary hypertension.
