摘 要:目的制备聚己内酯/聚三亚甲基碳酸酯(PCL/PTMC)静电纺丝支架材料,并研究其理化特征和组织相容性。方法采用静电纺丝技术制备PCL和不同配比的PCL/PTMC支架材料(3∶1,1∶1,1∶3,w/w);采用扫描电镜、傅里叶红外光谱、热重分析、差热分析及单轴拉伸试验检测已制备支架材料的形态特征、化学组成、热稳定性及力学性能;小鼠皮下组织植入各组实验材料,观察异物反应、材料降解及细胞在材料内生长情况。结果扫描电镜观察显示,PCL/PTMC(3∶1)、PCL/PTMC(1∶1)与PCL支架材料具有相似的纤维多孔性结构,PCL/PTMC(1∶3)支架材料发生纤维融合,失去纤维多孔性结构。傅里叶红外光谱显示,PCL/PTMC支架材料中PCL与PTMC分子间作用较弱,特征峰无明显改变。热重和差热分析显示,PCL/PTMC支架材料的热稳定性介于PCL和PTMC之间,随着PTMC含量的增加而降低。单轴拉伸试验显示,PTMC含量与材料的形态特征是影响PCL/PTMC支架材料力学性能的主要因素。小鼠皮下组织埋置实验显示,PCL、PCL/PTMC(3∶1)、PCL/PTMC(1∶1)材料植入2月后,宿主细胞侵蚀厚度分别为(23.85±3.71)%,(26.83±3.71)%和(29.15±2.33)%(P<0.05),PCL/PTMC(1∶3)材料表面可见侵蚀性降解,宿主细胞难以在材料内部生长。结论 PCL/PTMC静电纺丝支架材料的理化性质随材料中PTMC含量的增加发生规律性变化,纤维多孔性PCL/PTMC支架材料具有良好的组织相容性。Objective To prepare the electrospun polycaprolactone( PCL)/poly( trimethylene carbonate)( PTMC) scaffolds and investigate its characteristics and histocompatibility. Methods PCL and PCL/PTMC scaffolds( 3 ∶ 1,1 ∶ 1 and 1 ∶ 3,w/w) were prepared by electrospinning. The material characteristics,such as surface morphology,material composition,thermal behaviour and mechanical property,were investigated by scanning electron microscopy( SEM),fourier transform infrared( FTIR) spectra,thermal gravimetric analysis( TGA),differential scanning calorimetry( DSC) analysis and uniaxial tensile test,respectively. The foreign reaction,scaffold degradation and host cell growth in scaffold were observed in subcutaneous space of mice by implantation tests. Results SEM images showed that PCL/PTMC( 3 ∶ 1) and PCL/PTMC( 1 ∶ 1) scaffolds showed the micro/nano-fibres and porosity structure,similar with PCL scaffolds. Fibre fusion was found and the porosity structure was lost in PCL/PTMC( 1 ∶ 3) scaffolds. FTIR spectra showed that the molecular interaction between PCL and PTMC were weak and the characteristic peaks of PCL and PTMC changed slightly in PCL/PTMC scaffolds. TGA and DSC analysis showed that the thermal behaviour of PCL/PTMC scaffolds changed between PCL and PTMC,and the thermal stability decreased with the increasing content of PTMC. The uniaxial tensile tests showed that PTMC content and structure character of PCL/PTMC scaffolds were the main factors to affect the mechanical properties. After 2 months of implantation in subcutaneous space of mice,the cellular invasion ratio of PCL,PCL/PTMC( 3: 1) and PCL/PTMC( 1 ∶ 1) scaffolds was( 23. 85 ± 3. 71) %,( 26. 83 ± 3. 71) % and( 29. 15 ± 2. 33) %,respectively( P〈0. 05). In PCL/PTMC( 1 ∶ 3) group,there was obvious surface erosion and the host cells were difficult to grow into the scaffolds.
