摘 要:目的:研究肉桂灌胃给药后大鼠体内过程变化差异,为肉桂临床合理用药提供理论依据。方法:肉桂提取物不同剂量灌胃给药,采用HPLC法检测血浆及各脏器组织色谱峰,计算血浆样本与组织样本的总量统计矩参数如总量零阶矩(AUCT)、总量一阶矩(MCRTT)与总量二阶矩(VCRTT)。结果:经化学对照品对照,血浆样本与各脏器组织样本中均有马尿酸与肉桂酸色谱峰,血浆样本中马尿酸色谱峰随着剂量的增加而增大,随着给药时间的延长而增大,总量零阶矩AUCT随着给药剂量的增大而增大;组织样本中马尿酸在肾组织中的色谱峰最大;组织中的总量零阶矩AUCT肝肾>AUCT脾肺>AUCT心脑。结论:总量统计矩参数可评价肉桂灌胃给药大鼠机体后的体内过程变化差异,为肉桂的药动学研究与临床应用提供参考依据。Objective: To study the difference of the internal process of the components in rats after oral administration with extraction of Cinnamomum cassia,and to provide the theoretical basis for the rational use in clinic. Methods: Cinnamomum cassia extract was administered at different doses to rats,HPLC method was used to detect the plasma and tissue peaks of various organs,the total statistical moment parameters such as area under curve of total quantum(AUCT),mean chromatographic retention time of total quantum(MCRTT) and variance of chromatographic retention time of total quantum(VCRTT) in rats were calculated after treating different doses of extract. Results: Both hippuric acid and cinnamic acid was existed in plasma samples and tissue samples. Hippuric acid peak in plasma samples increased with the increasing of the dosage and the prolongation of medication,and AUCTincreased with the increasing of the dosage. The chromatographic peak of hippuric acid in kidney tissue was maximum. AUCTin liver and kidney tissues were larger than those in spleen and lung tissues,and those of heart and brain tissues were lowest,AUCT(liver kidney)AUCT(spleen lung)AUCT(heart brain).Conclusion: The total statistical moment parameters can be used to evaluate the internal process of the components in rats after oral administration with Cinnamomum cassia extract,which can provide a reference for the pharmacokinetic study and clinical application of Cinnamomum cassia.
