摘  要:R_2R_3-MYB转录因子在植物花青素合成过程中具有重要作用。本研究基于玫瑰转录组数据,克隆了一个玫瑰花青素相关R_2R_3-MYB基因,命名为Rr MYB10并对其进行生物信息学分析,期望通过克隆及分析探究MYB基因与玫瑰花瓣成色的关系,为改良玫瑰花色等奠定良好的基础。以‘紫枝’玫瑰(Rosa rugosa‘Zizhi’)为材料,通过RT-PCR以及RACE技术获得了该基因的c DNA全长。经过生物信息学分析,该基因全长871bp,开放阅读框699bp,编码232个氨基酸,其蛋白的分子式C1230H1904N348O356S6,相对分子质量为27455.13Da,等电点p I为9.01,该蛋白不稳定系数为42.31,属于不稳定蛋白。多重比对及序列分析发现其具MYB的保守域,系统进化树分析表明该基因与同科草莓等亲缘关系最近,与不同科物种亲缘关系远。R2R3-MYB transcription factors play a pivotal role in plant anthocyanin biosynthesis. Basing on the trascriptome of rosa rugose, we cloned one R2R3-MYB transcription factor related to anthocyanin biosynthesis, Rr MYB10,and conducted it's bioinformation analysis. By cloning and analyzing MYB transcription factor to study it's function in floral color, and to lay a good foundation for changing rosa rugosa's color.In this work,a full length c DNA sequence of MYB gene was obtained from petals of Rosa rugosa‘Zizhi'using RT-PCR and RACE.With the bioinformatics analysis,the cloned MYB gene was871 bp inthe full length,containing699 bp opening reading frame(ORF),which encoded 232 amion acids,the predicted MYB protein's molecular formula was C1230 H1904 N348 O356 S6,whose relative molecular mass was 27455.13 Da and isoelectric point was 9.01.The MYB protein's instability index was computed to be 42.31,which classified the protein as unstable.Through multiple alignment and sequence analysis, we found that it has conserved domains of MYB. Phylogenetic tree analysis showed that the gene was closely related to the same species of strawberry, and was far from different families.
