葛根芩连汤对小鼠血清及粪便代谢的影响

摘　　要：为了探索葛根芩连汤对小鼠血清色氨酸及主要代谢产物和粪便中部分短链脂肪酸的影响,试验通过葛根芩连汤灌胃小鼠,分别在干预7 d和14 d后采集样本,运用高效液相色谱-紫外检测器（HPLC-UVD）法测定小鼠血清中犬尿氨酸（Kyn）、色氨酸（Trp）与5-羟色胺（5-HT）浓度,气相色谱（GC）法测定小鼠粪便中乙酸、丙酸和丁酸含量。结果表明：与空白组相比,用药组小鼠血清Kyn和5-HT浓度明显升高（P〈0.05）,Trp无明显变化（P〉0.05）;给药7 d后粪便中丙酸浓度上升（P〈0.05）,用药14 d后乙酸、丙酸和丁酸浓度极显著升高（P〈0.01）。说明葛根芩连汤对小鼠血清Trp及主要代谢产物和粪便中部分短链脂肪酸存在一定的影响。In order to investigate the effect of Gegen Qinlian Decoction （GQD） on tryptophan and its major metabolites in sera, and on part of short chain fatty acids in feces in mice, GQD was given to mice by intragastric administration in this experiment, and samples were collected on day 7 and 14 post-treatment, respectively. The concentrations of kynurenine （ Kyn ）, tryptophan （ Trp ） and 5-hydroxytryptamine （ 5-HT ） in mouse sera were determined by HPLC-UVD, and the contents of acetic acid, propionic acid and butyric acid in mouse feces were measured by gas chromatography（ GC ）. The results showed that the concentrations of Kyn and 5-HT in treatment group increased significantly（ P〈0.05 ） and the content of Trp had no significant difference （P〉0.05）compared with those in control group;the concentration of propionic acid rose significantly （P〈0. 05）on day 7 post-treatment, and the concentrations of acetic acid, propionic acid and butyric acid increased very significantly （P〈0. 01 ） on day 14 post-treatment in treatment group compared with those in control group. The results suggested that GQD could have a certain effect on tryptophan and its major metabolites in sera,and on part of short chain fatty acids in feces in mice.
