摘  要:目的探讨WWOX基因对Tca8113细胞生长的影响及其作用机制。方法在脂质体Lipofectamine 2000的介导下,将携带WWOX基因的重组质粒WWOX-pc DNA3.0及空载体pc DNA3.0分别转染至Tca8113细胞中(Tca8113/WWOX组和Tca8113/vector组),同时设未转染组(Tca8113组),采用q PCR和Western blot法检测WWOX基因在转染细胞中的表达;MTT法检测Tca8113细胞的增殖能力;克隆形成试验检测Tca8113细胞的克隆形成能力;流式细胞术检测细胞凋亡情况;q PCR和Western blot法检测Tca8113细胞中肿瘤相关基因BCL2绑定组件3(BCL2binding component 3,BBC3)、叉头框蛋白O1(forkhead in rhabdom-yosarcoma,FOXO1)、凝血酶敏感素1(thrombospondin1,THBS1)及白介素-6(interleukin-6,IL-6)基因m RNA和蛋白的表达。结果与Tca8113/vector组和Tca8113组比较,Tca8113/WWOX组细胞中WWOX基因的m RNA和蛋白表达水平均升高,细胞增殖速度明显减慢(P<0.001),形成克隆数明显减少(P<0.001),细胞凋亡率明显升高(P<0.001),细胞中BBC3和FOXO1基因的m RNA和蛋白表达水平升高,IL-6和THBS1基因的m RNA和蛋白表达水平降低。结论 WWOX基因的过表达抑制了Tca8113细胞的生长,同时促进了Tca8113细胞的凋亡,可能与调节肿瘤相关基因BBC3、FOXO1、IL-6和THBS1的表达有关。Objective To investigate the effect of WWOX gene on growth of Tca8113 cells and the relevant mechanism.Methods Tca8113 cells were transfected with recombinant plasmid WWOX-pc DNA3. 0 carrying the full-length c DNA fragment of WWOX gene,using those transfected with empty vector and those untransfected as controls. The expression of WWOX gene in transfected cells was determined by q PCR and Western blot. The proliferation ability of Tca8113 cells was determined by MTT assay,while the clone formation ability by clone formation test,and the apoptosis by flow cytometry.The expressions of m RNA and protein of tumor-associated gene BCL 2 binding component 3(BBC3),forkhead in rhabdomyosarcoma(FOXO1),thrombospondin 1(THBS1) and interleukin-6(IL-6) were determined by q PCR and Western blot. Results Both the m RNA and protein expression levels of WWOX gene in Tca8113 cells transfected with recombinant plasmid WWOX-pc DNA3. 0 increased as compared with those in control groups,while the cell proliferation rate decreased significantly(P〈0. 001),the clones formed decreased signifcantly(P〈0. 001),and the cell apoptosis rate increased significantly(P〈0. 001). Both the m RNA and protein expression levels of BBC3 and FOXO1 increased,while those of IL-6 and THBS1 decreased. Conclusion The overexpression of WWOX gene inhibited the growth and promoted the apoptosis of Tca8113 cells,of which the mechanism might be associated with the regulation of tumorassociated genes BBC3,FOXO1,IL-6 and THBS1.
