摘  要:目的探讨柴胡疏肝散(CSS)对抑郁模型大鼠行为学及Notch1信号通路的影响。方法选择行为学评分相近的32只SD大鼠,分为对照组(CON)、模型组(CUMS)、阳性对照组(FLU)和干预组(CSS)。给予慢性不可预见性温和应激建立CUMS抑郁模型,采用糖水消耗和强迫抑郁进行行为学评估;免疫荧光检测大鼠海马神经元细胞的增殖;同时实时定量聚合酶链反应(Real-time PCR)和蛋白免疫印迹法(Western Blot)检测Notchl信号通路各因子(Notch1、Hes1、Hes5和Jagged1)的mRNA和蛋白表达水平。结果与CON组相比,CUMS组大鼠糖水偏爱百分比、游泳时间显著降低,差异有统计学意义(均P〈0.05)。与CUMS组相比,CSS组大鼠糖水偏爱百分比、游泳时间显著增加,均差异有统计学意义(均P〈0.05)。与CON组相比,CUMS组、FLU组及CSS组大鼠游泳不动时间显著增加,均差异有统计学意义(均P〈0.01)。与CUMS组相比,CSS组大鼠游泳不动时间显著减少,差异有统计学意义(P〈0.01)。与CON组[(750.00±27.51)个/mm2]相比,CUMS组[(338.75±29.61)个/mm2]、FLU组[(545.00±17.73)个/mm2]及CSS组[(529.38±13.74)个/mm2]大鼠海马齿状回颗粒细胞下层或颗粒细胞下层一颗粒细胞层BrdU阳性细胞数显著降低,均差异有统计学意义(均P〈0.01)。与CUMS组[(338.75±29.61)个/mm2]相比,CSS组大鼠海马齿状回颗粒细胞下层或颗粒细胞下层一颗粒细胞层BrdU阳性细胞数[(529.38±13.74)个/mm2]显著增加,均差异有统计学意义(P〈0.01)。与CON组相比,CUMS组大鼠海马Notch1、Hes1、Hes5和Jagged1mRNA和蛋白的表达水平显著降低,均差异有统计学意义(P〈0.05或P〈0.01);与CUMS组相比,FLU组和CSS组大鼠海马Notch1、hes1、hes5和jagged1mRNA和蛋白的表达水平显著增加,均差异有统计学意义(P〈0.05或Objective To investigate the effect of chaihu-shugan-san (CSS) on the behaviors and Notchl signal pathway in depression model rats. Methods Thirty-two SD rats with similar behavioral scores were divided into control group (CON), model group (CUMS),positive control group (FLU) and intervention group (CSS).The depression model was established by stimulating with chronic unpredictable mild stress (CUMS) , and the behaviors evaluation was assessed by sugar water consumption and forced depression.hnmunofluorescence was used to detect the proliferation of hippocampus neurons in rats, at the same time, real-time PCR and Western blot were used to detect the mRNA and protein expression levels of each factor (Notch1 ,Hes1 ,Hes5 and Jagged1) of Notchl signal pathway respectively. Results Compared with CON group, the percentage of sugar water preference and swimming length of rats decreased significantly in CUMS group (P〈0.05 and P〈 0.01 ). Compared with CUMS group, the percentage of sugar water preference and swimming length of rats increased significantly in CSS group(P〈0.05 ).Compared with CON group ,there was a significant increase in the inactivity length of rats between CUMS group, FLU group and CSS group, and the difference was statistically significant (P〈0.01).Compared with CUMS group ,the swimming length of rats in CSS group was significantly reduced, and the difference was statistically significant (P〈0.01).Compared with CON group( ( 750.00±27.51 )/mm2 ), the number of BrdU positive cells in the substratum or granuloeyte layer of the hippocampus dentate gyms of rats in CUMS group ( (338.75±29.61)/mm2) ,FLU group ( (54-5.00 ±17.73)/mm2) and CSS group ( ( 529.38± 13.74)/mm2 ) was significantly reduced(P〈0.01 ).Compared with CUMS group ((338.75±29.61)/mm2) ,there was a significant increase in the number of BrdU positive cells in the substratum or granulocyte layer of the hippocampus dentate gyms of rats in CSS gro
