摘  要:本研究克隆并鉴定香蕉枯萎病菌4号生理小种(Foc4)β1-微管蛋白(β1-tub)基因,采用Split-marker同源重组技术获得Foc4的β1-tub基因敲除突变体,通过测定突变体菌株的生长速度、产孢量、致病力及对多菌灵敏感性,研究β1-tub基因在香蕉枯萎病菌的生长发育及对多菌灵抗药性中的作用。结果表明:与Foc4野生型菌株相比,敲除突变体生长缓慢、菌丝畸形及产孢量增加,对巴西蕉苗的致病力明显减弱,但对多菌灵的抗性水平无明显变化。由此推测β1-tub基因可能在Foc4的生长发育、产孢及致病力等方面具有重要的作用。In this study, the β1-tubulin(β1-tub) gene in F.oxysporum f. sp. cubense race 4(Foc4) was cloned and identified. The gene-knockout mutants of β1-tub gene in Foc4 were obtained by split-marker homologous recombination technique. The growth rate, sporulation, pathogenicity and sensitivity to carbendazim of the mutant strain were studied to investigate the effects of β1-tub on the growth, development and carbendazim resistance in Foc4. The results demonstrated that the knockout mutants of β1-tub showed slow growth, the hyphal deformity and increased conidium production, and significantly decreased pathogenicity to banana(Cavendish, AAA), but no significant change in the resistance level to carbendazim when compared with the wild type isolate of Foc4. Therefore, we hypothesized that the β1-tub gene might play an important role in the growth, sporulation and pathogenicity of Foc4.
