摘 要:目的探讨瑞芬太尼对脑缺血再灌注大鼠学习和记忆功能的影响。方法选取成年雄性SD大鼠55只随机分为假手术组,模型组(脑缺血再灌注组),瑞芬太尼低剂量组、中剂量组、高剂量组,每组11只,制作大鼠全脑缺血再灌注模型,在大鼠全脑缺血再灌注后,低、中、高剂量组大鼠分别经尾静脉泵注瑞芬太尼5 min(速率分别为0.2,0.8,1.8μg·kg-1·min-1)。Morris水迷宫实验测试大鼠空间学习及记忆能力。HE染色观察大鼠海马CA1区神经元形态变化及存活的椎体细胞密度;免疫组化法测定海马CA1区的胆碱乙酰化转移酶(Ch AT)的含量。结果与假手术组比较,模型组、瑞芬太尼低剂量组逃避潜伏期明显延长,穿台次数、在平台象限内游泳的路径占总游泳路径的比例均明显减少(P<0.05);与模型组比较,瑞芬太尼中、高剂量组逃避潜伏期明显缩短,穿台次数、在平台象限内游泳的路径占总游泳路径的比例均明显增多(P<0.05);与假手术组比较,模型组、瑞芬太尼低剂量组椎体细胞及Ch AT阳性细胞数目明显减少,差异有统计学意义(P<0.05),瑞芬太尼中、高剂量组椎体细胞及Ch AT阳性细胞数目差异无统计学意义(P>0.05);与模型组比较,瑞芬太尼低剂量组椎体细胞及Ch AT阳性细胞数目差异均无统计学意义(P>0.05),瑞芬太尼中、高剂量组椎体细胞及Ch AT阳性细胞数目明显增多,差异均有统计学意义(P<0.05)。结论全脑缺血再灌注后大鼠经合适剂量的瑞芬太尼后处理,能明显提高海马CA1区的椎体细胞及Ch AT免疫阳性细胞的数量,提高全脑缺血再灌注模型大鼠的学习和记忆能力。OBJECTIVE To investigate the effects of remifentanil on learning and memory function in rats with cerebral ischemia reperfusion. METHODS Fifty-five healthy adult male rats were randomly divided into 5 groups ( 11 in each group ) : sham operation group, model group( whole brain ischemia reperfusion group) , remifentanil low dose, middle dose and high dose groups. The rat model of global cerebral ischemia-reperfusion was established, the rats in the low, middle and high dose group were injected with remifentanil 5 rain by the tail vein pump (the drip rate was 0.2,0.8,1.8 μg· kg-1. min-1, respectively). The spatial learning and memory ability were accessed by Morris water maze experiment. HE staining was used to observe the morphological changes of neurons in the hippocampus CA1 area and the viability of the vertebral body density. The content of ChAT in hippocampus of hippocampus was measured by immunohistochemistry. RESULTS Compared with the sham operation group, the escape latency was prolonged, while the number of times, the proportion of swimming paths in the platform quadrant were significantly reduced in the model group and low dose group (P〈 0.05). Compared with the model group, the escape latency was significantly shortened, while the number of times in the platform, the proportion of the swimming paths in the platform quadrant was significantly increased in the middle and high dose group (P〈0.05) , Compared with the sham operation group ,the number of vertebral cells and ChAT positive cells was statistically reduced in model group and low dose group (P〈0.05) ; with no significant difference in the middle dose group and the high dose group(P〉0.05). Compared with the model group ,the number of vertebral cells and ChAT positive cells increased significantly in the middle dose group and the high dose group (P〈0.05).
