白及化学成分分离鉴定

摘　　要：目的：基于白及提取物具有广泛的生物活性,本研究对白及95%乙醇提取物乙酸乙酯部位的化学成分进行研究,为白及的质量控制和开发利用提供一定的理论依据。方法：采用反复硅胶LH-20羟丙基葡聚糖（Sephadex LH-20）凝胶、制备薄层色谱、制备高效液相色谱等多种方法对白及的乙酸乙酯部位进行分离纯化,应用TLC,HPLC跟踪监测,分离得到纯度较高的单体化合物。根据理化性质及质谱（MS）,^1H-NMR和^13C-NMR等波谱数据鉴定化合物结构。结果：从白及的乙酸乙酯萃取部位分离得到12个化合物,分别鉴定为大黄素甲醚（1）,红灰青素（2）,8-C-对羟基苄基山柰酚（3）,N-苯甲酰-L-苯丙氨酸-N-苯甲酰-L-苯丙氨酸酯（4）,松脂醇（5）,（E）-4-羟基苯乙基3-（4＇-羟基苯基）丙烯酸酯（6）,对甲氧基二氢肉桂酸（7）,对羟基苯甲醛（8）,对羟基苯甲酸（9）,香草酸（10）,邻苯二甲酸二丁酯（11）,棕榈酸乙酯（12）。化合物类型包括蒽醌类（1和2）,黄酮类（3）,氨基酸衍生物（4）,木脂素（5）,苯丙素（6和7）,以及其他小分子芳香族化合物（8~11）和脂肪族（12）。结论：化合物2,3,4,6,7,11和12均为首次从白及植物中分离得到。Objective： To study the chemical constituents from the tubers of Bletilla striata. Method： The chemical constituents were isolated and purified by silica,Sephadex LH-20,and preparation of TLC. Their structures were identified by physicochemical properties and spectroscopic methods. Result： Twelve compounds were isolated from the ethyl acetate extracts of B. striata,including physcion（ 1）,erythroglaucin（ 2）,8-C-p-hydoxybenzylkaempferol（ 3）,N-benzoyl-L-phenylalaninyl-N-benzoyl-L-phenylalaninate（ 4）,pinoresinol（ 5）,（ E）-4＇＇＇-hydroxyphenethyl 3-（ 4＇-hydroxyphenyl） acrylate（ 6）, dihydo-p-methoxy cinnamic acid（ 7）, p-hydroxybenzaldehyde（ 8）, phydroxybenzoic acid（ 9）,vanillic acid（ 10）,dibutyl phthalate（ 11）,and ethyl palmitate（ 12）. The types of these compounds included anthraquinones（ 1 and 2）,flavonoid（ 3）,amino acid derivative（ 4）,lignan（ 5）,phenyl propanoids（ 6 and 7）,other aromatic compounds（ 8-11） and aliphatic compound（ 12）. Conclusion： Compounds 2,3,4,6,7,11 and 12 were reported from B. striata for the first time.
