摘 要:目的:探讨苔黑酚葡萄糖苷(Orcinol Glucoside,OG)对甲基苯丙胺(Methamphetamine,MA)引起的焦虑样行为的影响以及对海马区多巴胺、高香草酸、5-羟色胺、5-羟吲哚乙酸的影响。方法:通过高架十字迷宫实验研究OG对MA(1 mg/kg)诱导的焦虑样行为影响;通过小鼠自发活动实验观察OG对MA诱发的小鼠高活动量的影响;采用高效液相质谱联用技术检测小鼠脑内多巴胺和5-羟色胺及其代谢物含量。结果:OG能够显著性增加MA处理小鼠进入高架十字迷宫开放臂时间,降低MA小鼠的自发活动;神经化学检测发现MA组小鼠脑内海马区多巴胺、5-羟色胺及髙香草酸含量明显增加;而OG能够显著性降低MA小鼠脑内高香草酸含量,同时有降低MA诱导的多巴胺及5-羟色胺水平增加的趋势。结论:OG能够有效减轻MA引起的焦虑样行为,且对MA导致的脑内高香草酸水平有回调作用,可作为对抗MA精神症状的潜在治疗药物。Objective: To investigate the effects of orcinol glucoside(OG) on anxiety-like behaviors induced by methamphetamine( MA) and its effects on dopamine and dopamine metabolite-homovanillic acid( HVA).Methods: The effects of OG on the anxiety-like behaviors in mice induced by methamphetamine( 1 mg/kg) were measured by the elevated plus-maze test. The content of dopamine and HVA in the mouse brain were detected by liquid chromatography-mass spectrometry.Results: MA significantly decreased the time that mice spent in the open arms,indicating MA was anxiogenic. OG significantly reversed the decrease of the time that mice spent in the open arms and the increase of locomotion induced by MA. Neurochemical detection revealed that the content of HVA in the hippocampus of mice treated by MA was increased compared to that in the control group; OG significantly decreased the content of HVA in the brain of mice treated by MA.Conclusion: OG can effectively reduce the anxiety-like behavior induced by MA in mice,and reverse the high level of HVA in the brain of MA-treated mice. Therefore,OG may be a potential therapeutic drug against MA-induced mental disorders.
