摘　　要：目的分析河南省登封市2014-2016年30株布鲁氏菌的生物型别及PFGE分子分型特征,为布鲁氏菌病的病原学监测,暴发预警、溯源提供基线数据。方法采集试管凝集试验(SAT)阳性病人静脉血10ml,双相血培养瓶培养与生化鉴定,AMOS-PCR鉴定布鲁氏菌生物型别;阳性菌株利用脉冲场凝胶电泳(PFGE)和BioNumerics7.0软件进行指纹图谱分析。结果阳性病例以青壮年男性职业和密接人群为主;30株布鲁氏菌经鉴定28株为羊种,2株为牛种。羊种布鲁氏菌经脉冲场凝胶电泳后可分为2种带型,带型相似度在97.7%～100%之间;牛种布鲁氏菌可分为2种带型,带型相似度96.3～100%。结论河南省监测哨点医院人感染布鲁氏菌存在羊种和牛种两种型别,羊种为优势型别;羊种与牛种菌株在PFGE带型特征上存在较大差异度。Objective To identify and analyze Brucella strains biotypes and molecular typing characteristics of PFGE in Dengfeng city of Henan Province from 2014 to 2016,and to provide baseline data for pathogen monitoring,outbreak warning and traceability of brucellosis. Methods 10 ml SAT test positive patients ＇ blood was collected and cultured with double-phase blood bottle. The positive strain＇s biotypes were identified by AMOS-PCR. Morphological and biochemical identification,as well as fingerprint characters,were analyzed by pulsed-field gel electrophoresis（ PFGE） and BioNumerics7. 0 software. Results The positive cases were mainly occupational and close contacts with young male adults. 30 strains of Brucella were identified by morphological,biochemical and AMOS-PCR methods,in which 28 strains were sheep species,and 2 strains were cattle species. PFGE results indicated that the similarity coefficient among 28 sheep species was 97. 7%-100% with 2 belt types,and the similarity coefficient between 2 cattle species was 96. 3%-100% with 2 belt types. Conclusion There are sheep and cattle species of Brucella isolated from patients in sentinel hospital of Dengfeng city. Sheep species is the main biotype,and there is a great difference in PFGE band type between sheep and cattle strains.
