精子细胞XRCC1基因rs25487位点多态性与少弱精患者的相关性研究

摘　　要：目的探究精子细胞X射线交叉互补修复基因(XRCC1)rs25487位点的多态性及与少弱精的相关性。方法收集182例男性少弱精患者和73例正常男性的精液,进行精液常规分析,包括精液量、液化时间、精液浓度及精子活力,然后采用聚合酶链反应-限制性段长度多态性(PCR-RFLP)方法对精子细胞中的XRCC1基因进行分型。检测XRCC1基因rs25487位点的基因分型和等位基因频率,分析其与少弱精症的相关性。结果少弱精患者的精子浓度和精子活力显著降低,液化时间显著延长。与GG基因型相比,携带AA基因型个体患少弱精症的风险是GG基因型的7.84倍(95%CI:1.019~60.365)。结论精子细胞XRCC1基因rs25487位点AA基因型是男性患少弱精症的危险因素。Objective To explore the association between X-ray repair cross complementing group 1（XRCC1）gene rs25487 locus polymorphisms in sperm DNA of oligoasthenotspermia.Methods 182 oligoasthenotspermia patients and 73 healthy male were recruited in this study.Their semens were collected once semen routine analysis had done.Then,we use polymerase chain reaction-restriction fragment length polymorphism（PCR-RFLP）method to genotype the polymorphism of XRCC1 gene rs25487 locos.Results Compare with the healthy control group,the sperm concentration and motility were significantly decreased and the liquefaction were significantly prolonged in the oligozoospermia or azoospermia patients.AA genotype,compare with GG genotype,demonstrated a significantly increased risk for oligozoospermia or azoospermia patients（OR=7.84;95%CI=1.019～60.365）.Conclusion Our results demonstrated that AA genotypes in XRCC1 gene rs25478 locus may be a risk factor for oligozoospermia or azoospermia patients.
