摘  要:目的探索亚型为CRF65_CPX艾滋病病毒(HIV)毒株在中国不同高危人群间的传播特征,基因序列及其CTL表位的变化。方法收集2017年6月之前中国鉴定HIV-1亚型为CRF65_CPX的毒株的pol基因区序列,使用比对好的pol基因区序列进行贝叶斯分析。以系统进化树为根据,选择其中同性性传播人群和异性性传播人群序列,分别生成共享序列后,比较两个人群氨基酸位点和HLA-B位点等位基因限制的CTL表位的差异。结果共收集获得CRF65_CPX HIVpol基因区序列52条。贝叶斯分析结果显示,异性性传播人群的最近共同祖先的时间为1999年,同性性传播人群的最近共同祖先的时间为2003年,CRF65_CPX毒株出现在异性性传播人群的时间早于同性性传播人群。与异性性传播人群相比,同性性传播人群pol区共享序列有10个氨基酸位点发生变化;丢失了两个有数据证明的保护性CTL表位(B*2702、B*5103)。结论 CRF65_CPX毒株为中国近些年才发现的多重重组的新型毒株,其在中国的传播途径从异性性传播转变为以同性性传播为主,氨基酸位点和CTL表位在传播过程中也产生了变化,这种变化可能加速了疾病进程,故应引起并加强对该亚型及其类似的新型病毒株不断扩散传播的重视。Objective To explore the transmission characteristics,consensus sequences and CTL epitopes of CRF65_CPX in different high-risk population in China.Methods We collected the pol region sequences of CRF65_CPX before June 2017 in China.Bayesian analysis was performed by using aligned pol region sequences.Based on the phylogenetic tree,the homosexual and heterosexual sequences were selected to generate the consensus sequences respectively and to compare the difference of amino acid and CTL epitopes restricted by HLA-B in the two population.Results A total of 52 pol sequences were obtained in this study.Bayesian analysis showed that the time of the most recent common ancestor of heterosexual and homosexual population were inferred as 1999 and 2003 respectively.CRF65_CPX was present in heterosexual population earlier than in homosexual population.Compared with the heterosexual population,10 amino acid sites changed,and two validated protective CTL epitopes(B *2702,B *5103)were lost in homosexual population.Conclusion The risk exposure of CRF65_CPX has shifted from heterosexual transmission to homosexual in China.Some amino acid sites and CTL epitopes also change during the transmission process,which may accelerate the progression of the disease.Therefore,the spread of CRF65_CPX deserves attention.
