摘 要:目的:制定BY-1型全自动末梢血细胞混匀仪对血小板计数使用参数。方法:对30例健康供体末梢血用手工法和末梢血细胞混匀仪混匀法比较使用不同温度(10~37℃)和不同时间(10~160s)对血小板结果影响;采集30例健康供者、30例血小板高于参考区间的患者和30例血小板低于参考区间的患者末梢血分别用仪器法和手工法混匀计数血小板,并进行两者比较;每一样本均用静脉血为对照组,并将本研究结果与静脉血细胞分析结果进行比较。结果:将末梢血细胞混匀仪温度设定为30℃,然后分别混合10~160s,结果表明末梢血混匀时间在40~80s时,各指标比较稳定,与对照组结果相近;混匀时间在20s内结果不稳定,超过100s时结果明显高于对照组。将末梢血细胞混匀仪混匀时间设定为60s,然后在10~37℃状态下分别混匀并计数,混匀仓温度为10℃时血小板结果最低,与对照组比较差异有统计学意义(P<0.05);混匀仓温度在15~37℃范围对结果影响不明显,以30℃时结果最稳定。分别用仪器和手工法混匀后检测低值、高值和正常值各30例末梢血血小板样本,2种混匀方法计数的血小板结果,在组内之间比较大多数差异明显,各值组间比较差异有统计学意义(P<0.05)。结论:末梢血细胞混匀仪在设定最佳温度和混匀时间后混合的样本计数血小板结果准确性和稳定性明显好于手工混匀法。Objective:To develop the using parameters of BY-1 automatic peripheral blood cell mixers for platelet count.Method:Thirty patients with healthy donor peripheral blood were compared with those of peripheral blood cell mix method.The effects of different temperature(10-37℃)and different time(10-160 s)on platelet were compared,30 cases of platelets higher than the reference value of patients and 30 cases of platelets below the reference value of peripheral blood were used instrumental and manual method of mixing count platelets,and the results were compared;each sample were treated with venous blood and the results of this study was compare with the results of venous blood cell analysis.Result:The temperature of the peripheral blood cell mixture was set at 30℃,and then mixed for 10-160 s.The results showed that the peripheral blood was mixed at 40-80 s,the indexes were stable,and the results were similar to those of the control group.The mixing time was 20 s.The results were unstable,more than 100 swhen the results were significantly higher than those of the control group.The mixing time of the peripheral blood cell blender was set to 60 sand then mixed and counted at 10℃ to 37℃.The results showed that the platelet was the lowest when the mixing temperature was 10℃,which was significantly different from that of the control group(P〈0.05).The effect of mixing bin temperature in the range of 15-37℃was not obvious,and the results were the most stable at 30℃.The platelet samples were measured in 30 cases of low value,high value and normal value by instrumentation and manual method.The platelet outcome difference between group and group was obvious,and the difference between the two groups was statistical differences(P〈0.05).Conclusion:The accuracy and stability of the platelet count of the samples mixed with the samples after the optimal temperature and mixing time of the peripheral blood cell mixers were significantly better than those of the manual mixing method.
