摘 要:目的研究冰片对大鼠脑缺血再灌注损伤后超氧化物歧化酶(SOD)活性、炎症反应和血脑屏障通透性的作用及其机制。方法按照体重将大鼠随机分成3组,每组10只:假手术组、模型组和实验组。造模前,实验组给予10%冰片(0.5 g·kg^(-1))和石蜡油2 m L灌胃,1次/天;假手术组和模型组给予相同体积0.9%Na Cl,连续7 d。用颈总动脉夹闭法制备脑缺血-再灌注损伤模型。缺血0.5 h后再灌注24 h后,用黄嘌呤氧化酶法与分光光度法分别检测缺血脑组织SOD活性和髓过氧化物酶(MPO)活性和伊文思蓝(EB)含量;用酶联免疫吸附实验测定脑组织肿瘤坏死因子-α(TNF-α)含量。结果脑缺血-再灌注24h后,假手术组、模型组和实验组得大鼠脑组织SOD活性分别是(332.76±3.30),263.17±1.47),(286.01±2.34)U·mg-1;这3组的大鼠脑组织MPO活性分别是(0.17±0.02),(0.96±0.15),(0.68±0.11)U·g-1,模型组与正常组比较,差异有统计学意义(P<0.01);实验组与模型组比较,差异均有统计学意义(P<0.01)。假手术组、模型组和实验组的大鼠脑组织TNF-α含量分别是(0.36±0.05),(0.87±0.02),(0.69±0.24)U·g-1,模型组与正常组比较,差异有统计学意义(P<0.01);实验组与模型组比较,差异有统计学意义(P<0.01)。结论冰片对脑缺血再灌注损伤模型大鼠的脑组织具有神经保护作用。Objective To explore the effects of borneol on superoxide dismutase( SOD) activity,inflammatory reaction and permeability of blood-brain barrier in rats with cerebral ischemia-reperfusion injury and their potential mechanisms. Methods Thirty rats were divided into sham operation group,model group and experimental group. The experimental group was preoperatively administered,10% borneol( 0. 5 g·kg-1)paraffin oil 2 m L was gavagedfor each day,for 7 days. The sham group and model group were given the same volume of 0. 9% sodium chloride injection. Cerebral ischemia-reperfusion model was established with carotid artery clamping method. SOD activity,myeloperoxidase( MPO)activity and the contents of Evans blue( EB) in rat brain were measured at the 24 h after the cerebral reperfusion post 0. 5 h ischemia. Contents of tumor necrosis factor-α( TNF-α) in rat brain were detected by enzyme linked immunosorbent assay. Results The SOD activity insham operation group,model group and experimental group were( 332. 76 ± 3. 30),263. 17 ± 1. 47),( 286. 01 ± 2. 34)U·mg-1,respectively. The MPO activity in the 3 groups were( 0. 17 ± 0. 02),( 0. 96 ± 0. 15),( 0. 68 ± 0. 11)U·g-1,respectively. The TNF-α content in the 3 groups were( 0. 36 ± 0. 05),( 0. 87 ± 0. 02),( 0. 69 ± 0. 24)U·g-1,respectively. Comparison between the normal group with the model group,the differences of the three factors were statistically significant( all P〈0. 01). Comparison between the model group with the experimental group,the difference of the three factors were statistically significant( P〈0. 01). Conclusion Borneol exerts neuroprotective effects on cerebral ischemia-reperfusion injury in rats.
