摘  要:目的探讨隔药灸神阙穴治疗原发性痛经的可能作用机制。方法将40只SD雌性大鼠随机分为空白组、模型组、隔药灸组、灌胃组,每组10只。除空白组外,其余各组大鼠采用皮下注射苯甲酸雌二醇和催产素联用的方法制备原发性痛经大鼠模型。第5日开始,隔药灸组给予隔药灸神阙穴;灌胃组给予复方益母草膏溶液80 g/(kg·d)灌胃,两组均每日1次,连续6日。空白组皮下注射等容积生理盐水,每日1次,连续10日。观察各组大鼠腹腔注射催产素后30 min内的扭体反应情况。给药结束后检测大鼠腹主动脉血,进行主成分分析和正交偏最小二乘法-判别分析,寻找隔药灸组与灌胃组主要差异性代谢产物和代谢通道。结果与模型组比较,隔药灸组、灌胃组扭体潜伏期均延长,扭体次数均减少(P<0.01);与灌胃组比较,隔药灸组扭体潜伏期、扭体次数差异均无统计学意义(P>0.05)。隔药灸组筛选后得到10种差异性代谢物,其中鉴定了4种对分类有重要贡献的差异性代谢物,主要表现为血液中的羟脯氨酸、丙酮酸、L-半胱氨酸、谷氨酸的上升,涉及的代谢通路有谷胱甘肽代谢通道、氨基酸代谢通道、苯丙氨酸代谢通道。灌胃组筛选后得到12种差异性代谢物,其中鉴定了3种对分类有重要贡献的差异性代谢物,主要表现为血液中的氢化肉桂酸、硫磺酸、2-酮己二酸3的下降,涉及的代谢通路有氨基酸代谢通道、苯丙氨酸代谢通道。结论隔药灸神阙穴对通过谷胱甘肽代谢通道代谢的胶原蛋白类氨基酸的影响可能是其治疗原发性痛经的特异性机制之一。Objective To explore the possible mechanism of herbs-partition moxibustion at Shenque( CV8) acupoint in treating primary dysmenorrhea. Methods Forty female SD rats were randomly divided into blank group,model group,herb-partition moxibustion group,and intragastric administration group,with 10 rats in each group.Except for the blank group,rats were treated with subcutaneous injection of estradiol benzoate and oxytocin to prepare a primary dysmenorrhea rat model. On the 5 th day,the herb-partition moxibustion group was given moxibustion at Shenque( CV8) point; the intragastric administration group was given compound folicum roxburghii cream 80 g/( kg·d) to gavage. Both groups were given once a day for 6 consecutive days. The blank group was subcutaneously injected with equal volume of normal saline once a day for 10 consecutive days. The writhing response was observed within 30 min after intraperitoneal injection of oxytocin. Abdominal aorta blood was measured after the end of administration. Principal component analysis and orthogonal partial least squares discriminant analysis were performed to find the main metabolites and metabolic pathways in the herb-partition moxibustion group and the intragastric administration group.Results Compared with the model group,the writhing latency of the herb-partition moxibustion group and the gavage group both prolonged and the writhing times decreased( P〈0. 01). Compared with the gavage group,the writhing latency and writhing frequency of the herb-partition moxibustion group were different without statistical significance( P〉 0. 05). After screening the herb-partition moxibustion group,10 differential metabolites were obtained,among which4 differential metabolites that contributed to the classification were identified,mainly hydroxyproline,pyruvate,and L-cysteine in the blood.
