Effect of Qidan Fugan Granules(芪丹复感颗粒) on Intestinal Function in Low Respiratory Tract Mucosal Immunosuppressed Model Mice

摘  要:目的探讨芪丹复感颗粒治疗小儿反复呼吸道感染的可能作用机制。方法将60只昆明种小鼠随机分为模型组、空白对照组、匹多莫德组及芪丹高、中、低剂量组,每组10只。除空白对照组外,其余各组小鼠采用卵清蛋白致敏及雾化激发制备呼吸道黏膜免疫低下模型。造模后芪丹高、中、低剂量组分别给予芪丹复感颗粒0.324、0.162、0.081 g/10 g灌胃;匹多莫德组给予匹多莫德颗粒0.024 g/10 g灌胃;模型组及空白对照组给予0.1 ml/10 g生理盐水灌胃,每天1次。灌胃2周后,计算各组小鼠脾脏指数、胸腺指数、胃肠推进率,并测定分泌型免疫球蛋白A(sIgA)含量。结果模型组小鼠脾脏指数、胸腺指数、胃肠推进率、sIgA均低于空白对照组(P<0.05或P<0.01);匹多莫德组及芪丹高、中、低剂量组小鼠脾脏指数、胸腺指数、胃肠推进率及sIgA均高于模型组(P<0.05);芪丹高、中、低剂量组之间各指标比较差异均无统计学意义(P>0.05)。结论芪丹复感颗粒可以促进呼吸道黏膜免疫低下小鼠的肠蠕动,升高肠液sIgA水平,可能是其治疗小儿反复呼吸道感染的作用机制之一。Objective To explore the possible mechanism of Qidan Fugan Granules( 芪丹复感颗粒) in treating pediatric recurrent respiratory tract infection. Methods Sixty Kunming mice were randomized into blank control group,model group,pidotimod group,Qidan Fugan Granules of high,medium and low dose groups,with 10 mice in each. Except for the blank control group,mice in the other groups were sensitized with ovalbumin and aerosolized to prepare a model of respiratory mucosal immunity. After modeling,the high,medium,and low doses of Qidan Fugan Granules group were given 0. 324,0. 162,and 0. 081 g/10 g,respectively,while those in the pidotimod group were given 0. 024 g/10 g pidotimod Gandudu. The blank control group was given intragastric administration of 0. 1 ml/10 g saline once daily. After 2 weeks of gavage,the spleen index,thymus index,and gastrointestinal propulsive rate of each group of mice were calculated,and the content of secretory immunoglobulin A( s Ig A) was determined.Results The spleen index,thymus index,gastrointestinal propulsion rate,and s Ig A in the model group were lower than those in the control group( P 〈0. 05 or P 〈0. 01); the spleens index,thymus index,gastrointestinal propulsion rate,and s Ig A of the mice in the high-,middle-,and low-dose groups of Piedemold and Qidan Fugan Granules groups were all higher than those in the model group( P 〈0. 05). There were no significant differences in the indexes among the high,middle,and low doses of Qidan Fugan Granules group( P〉 0. 05). Conclusion Qidan Fugan Granules can promote intestinal peristalsis in mice with low respiratory mucosal immunity and increase the s Ig A level in intestinal fluids,which may be one of the mechanisms for the treatment of repeated respiratory infections in children.
