摘 要:目的研究辛麻颗粒对哮喘小鼠胸腺基质淋巴细胞生成素(thymic stromal lymphopoietin,TSLP)分泌及气道炎症的影响。方法将BALB/C雌性小鼠50只随机分为正常组、哮喘组、辛麻低剂量组、辛麻高剂量组、地塞米松组,每组10只。除正常组外,其余4组均由卵清白蛋白(OVA)致敏和激发建立小鼠慢性哮喘模型,造模时间共8周。辛麻低剂量组从第21天开始每天2次灌胃辛麻颗粒(1.3 g/kg),辛麻高剂量组的用药剂量是低剂量组的2倍,地塞米松组在每日雾化前腹腔注射地塞米松(10 mg/kg)。进行肺泡灌洗液细胞分类计数及IL-4和IL-13测定,HE染色观察气道炎症,测定肺组织TSLP蛋白。结果HE染色观察哮喘组气道炎症明显;哮喘组小鼠细胞总数、嗜酸粒细胞计数、淋巴细胞计数以及中性粒细胞计数较正常组升高(P<0.05),不同剂量辛麻组、地塞米松组较哮喘组下降(P<0.05),辛麻组低于地塞米松组(P<0.05),两种剂量辛麻组比较,差异无统计学意义(P>0.05);哮喘组肺泡灌洗液IL-4和IL-13浓度较正常组升高(P<0.05),两辛麻组、地塞米松组较哮喘组下降(P<0.05),两辛麻组均低于地塞米松组(P<0.05),不同剂量辛麻组比较,差异无统计学意义(P>0.05);哮喘小鼠TSLP蛋白表达较正常组增强(P<0.05),不同剂量辛麻组、地塞米松组较哮喘组下降,两辛麻组与地塞米松组比较,差异有统计学意义(P<0.05),不同剂量辛麻组比较,差异无统计学意义(P>0.05)。结论辛麻颗粒可能通过降低TSLP的表达,抑制Th2优势的免疫作用,从而改善了哮喘小鼠的气道炎症,但其作用与剂量无明显相关。Objective To observe the effect of Xinma Granule( XMG) on the secretion of thymic stromal lymphopoietin( TSLP) and airway inflammation of asmatic mice. Methods Fifty female BALB/C mice were randomly divided into 5 groups,i. e.,the normal group,the asthmatic group,low and high dose XMG groups,and the dexamethasone( TXM) treated group,10 in each group. In the latter four groups,chronic asthmatic models were established by ovalbumin( OVA) allergy and activation. From the 21 st day,XMG at 1. 3 g/kg was administrated to mice in the low dose XMG group by gastrogavage,twice per day. XMG at 2. 6 g/kg was administrated to mice in the high dose XMG group,twice per day. Dexamethasone( 10 mg/kg)was intraperitoneally injected to mice in the dexamethasone treated group everyday before atomization. The modeling lasted for 8 weeks. Cell counts in bronchoalveolar lavage fluid( BALF) were performed and classified. The levels of IL-4 and IL-13 in BALF were detected. The airway inflammation was evaluated by HE staining. TSLP protein in the lung tissue was determined.Results HE staining showed the inflammation was most obviously seen in the asthmatic group. The total cells,counts of eosinophils,lymphocytes,and neutrophilic granulocytes increased higher in the asthmatic group than in the normal group(P〈0. 05). They were all decreased more in the two XMG groups and the dexamethasone treated group than in the asthmatic group(P〈0. 05). They were lower in the two XMG groups than in the dexamethasone treated group(P〈0. 05). There were no statistically significant differences between the two XMG groups(P〈0. 05). The counts of IL-4 and IL-13 in BALF were higher in the asthmatic group than in the normal control group(P〈0. 05). They were all decreased more in the two XMG groups and the dexamethasone treated group than in the asthmatic group(P〈0. 05).
