摘 要:为研究湖北省内鸡传染性鼻炎病原菌的特性,本研究从湖北恩施某地鸡场疑似传染性鼻炎的病鸡样品中分离获得一株疑似副鸡禽杆菌(APG)的病原菌。通过细菌的分离培养、形态观察、生化试验、革兰氏染色、动物回归试验和PCR等方法对其进行鉴定。结果显示,分离的病原菌具有APG的形态特征和生化特性;16S rRNA序列比对分析与已报道的APG相应序列同源性高于98%;另外,利用特异PCR,分别检测到p250,pA14和pYMH5等3种质粒的特异基因片段;通过培养试验,该分离株能在无NAD的TSB(含鸡血清)中生长。这是国内首次发现无需NAD且同时存在3种质粒的APG。这一的发现警示国内的APG流行情况复杂,应引起畜牧兽医工作者的注意。Avibacterium paragallinarum is a causative agent of avian infectious coryza in chicken. To understand the characteristics of A.paragallinarum, a A.paragallinarum was isolated from chicken in Enshi, Hubei province, and identified by bacterial isolation and culture, morphological observation, biochemical tests, PCR and animal infection test. As results, the characteristics of the morphology, the biochemical tests and 16S rRNA sequence (more than 98% homology) of the isolate were identity to the A.paragallinarum reference strains. In addition, the isolate had the ability to grow in TSB medium containing chicken serum without NAD. Moreover, the 3 plasmids of p250, pAl4 and pYMH5 were detected in the isolate by PCR. This is the first report of NAD-independent and embodied the 3 plasmids of A.paragallinarum in China. Our results suggested that the A.paragallinarum infection was becoming more and more complex, which should be paid attention to by specialists.
