摘  要:为研究单核增生李斯特菌ICDC-LM188对阿洛糖的利用及阿洛糖对该菌株基因表达的影响,本研究利用含有不同碳源的MWB培养基和LB培养基分别进行单核增生李斯特菌在阿洛糖作用下的生长试验,采用RNA测序并进行主要差异表达基因分析。生长试验结果显示阿洛糖可以抑制ICDC-LM188生长。差异表达基因分析结果显示,5个上调表达基因均与已知的阿洛糖代谢相关,14个下调表达基因中部分与丙酮酸代谢相关,而丙酮酸代谢的抑制会影响菌体生长,推断丙酮酸代谢的抑制可能是阿洛糖抑制部分单核增生李斯特菌生长的原因。本研究为优化LAEB培养基提供了方向及对单增李斯特菌的糖代谢相关研究有借鉴意义。To study the effects of D-allose on the growth and differential expression genes of Listeria monocytogenes, MWB and LB mediums containing different carbon sources were utilized to detect the utlization of D-allose in L.monocytogenes strains ICDC-LM188. The growth curves of L.monocytogenes in different medium indicated that D-allose inhibited the growth of some L.monocytogenes. Here the parts of L.monocytogenes were designated as non-utilization L.monocytogenes. According to the analysis of differential expression genes in ICDC-LM188 in different LB mediums, five genes were found to be up-regulated and related to D-ribose metabolism, a total of 14 down-regulated genes were detected and associated with metabolism, motility and so on. Moreover, six of down-regulated genes were related to pyruvate metabolism. Inhibition of pyruvate metabolism was inferred to have a disadvantage action on bacteria growth. The study has provided the reference data for modification of LAEB medium and further research about the carbohydrate metabolism of L.monocytogenes.
