摘  要:目的初步探讨当归"润肠通便"功效的物质基础。方法将90只昆明种小鼠随机分为空白组,血虚便秘组,阳性对照组,当归高、低剂量组,当归多糖高、低剂量组,当归油高、低剂量组等9组。除空白组外,其余各组小鼠均通过皮下注射乙酰苯肼(APH)、腹腔注射环磷酰胺(CPA)联合灌服复方地芬诺酯片(DPN)混悬液建立血虚便秘模型,造模第14天开始,各组小鼠分别灌服生理盐水、常通舒颗粒溶液、当归水煎液、当归多糖水溶液、当归油乳剂,连续灌胃2周。观察各组小鼠一般情况及体质量变化,记录首粒黑便排出时间,测定血液学指标,并计算粪便和结肠含水量。结果与空白组比较,血虚便秘组小鼠基本能体现中医血虚便秘证候,外周血白细胞(WBC)、红细胞(RBC)及血红蛋白(HGB)、红细胞压积(HCT)均显著降低(P<0.05或P<0.01),排便时间显著延长(P<0.01),粪便和结肠含水量显著降低(P<0.01)。与血虚便秘组比较,当归水煎液可改善模型小鼠血虚便秘证候,显著升高小鼠外周血RBC、WBC、HGB和HCT(P<0.05或P<0.01),显著缩短排便时间(P<0.05或P<0.01),升高粪便和结肠含水量(P<0.05或P<0.01);当归多糖可改善模型小鼠血虚证候,显著升高模型小鼠外周血RBC、WBC、HGB和HCT(P<0.05),升高小鼠结肠含水量(P<0.05),但对排便时间无明显影响(P>0.05);当归油可改善模型小鼠便秘证候,升高粪便和结肠含水量(P<0.05),高剂量当归油可显著缩短排便时间(P<0.05),但对外周血RBC,WBC,HGB和HCT无明显影响(P>0.05)。结论当归多糖和当归油可能是当归发挥"润肠通便"功效的重要组分,但尚不能代表当归的整体效应。Objective To study the medicinal material base of Angelica sinensis in nourishing bowel to relieve constipation. Methods 90 mice were randomly divided into 9 groups: blank group,constipation group,positive control group,high-and low-dose angelica sinensis group,high-and low-dose angelica sinensis polysaccharide( ASP) group,high-and low-dose angelica volatile oil group. Except the blank group,mice of the rest groups were subcutaneously injected with acetylphenyhydrazine( APH) and intraperitoneally injected with cyclophosphamide( CPA) and orally administrated with diphenoxylate( DPN) to copy aninmal models with constipation due to blood deficiency. Day 14 of modelling,the mice were administered with saline,Changtongshu Gradule,angelica sinensis decoction,angelica sinensis polysaccharide and angelica volatile oil. Then the general status,body mass,defecation time,levels of RBC,WBC,HGB and HCT in peripheral blood,the water content in stool and colon of all groups were tested. Results Compared with blank group,the mice in consptipation group were in accordance with the syndrom of constipation due to blood deficiency,the levels of RBC,WBC,HGB,HCT in peripheral blood significantly decreased( P〈0.05 or P〈0.01),the defecation time was obviously prolonged( P 0.01),the water content of the stool and colon clearly decreased( P〈0.01). Compared with consptipation group,syndromes of blood deficiency and conspipation were improved in high-and low-dose Angelica sinensis groups showing the levels of RBC,WBC,HGB and HCT in peripheral blood significantly increased( P 0. 05 or P 0. 01),the defecation time was significantly shortened( P〈0.05 or P〈0.01),while the water content of colon and stool increased( P 0.05 or P 0.01). Syndrome of blood deficiency was improved in high-and low-dose ASP groups,showing the levels of RBC,WBC,HGB and HCT significantly increased( P〈0.05),the water content of colon increased( P〈0.05),but the defecation time had no change( P 0. 05)
