摘 要:目的研究黄芪多糖对支气管哮喘模型小鼠气道炎症及水通道蛋白5(AQP5)表达的影响。方法将60只雄性C57BL/6J小鼠随机分为空白组,模型组,地塞米松组,黄芪多糖低、中、高剂量组等6组,每组10只,除空白组外,其余各组通过腹腔注射卵清蛋白(OVA)与氢氧化铝配制的致敏液及OVA雾化吸入复制小鼠支气管哮喘模型,空白组、模型组予生理盐水0.2 m L灌胃,黄芪多糖低、中、高剂量组分别以100.0,200.0,300.0 mg/kg剂量灌胃黄芪多糖溶液,地塞米松组以0.8 mg/kg剂量腹腔注射地塞米松磷酸钠注射液。各组予相应药物干预30 d后,比较各组小鼠支气管肺泡灌洗液(BALF)细胞总数、分类计数及肺湿干重比值;采用酶联免疫法测定各组小鼠血清、BALF中白细胞介素-5(IL-5)、γ-干扰素(IFN-γ)的含量及AQP5的表达水平,并进行组间比较。结果与空白组比较,模型组小鼠BALF细胞总数,中性粒细胞、嗜酸性粒细胞、单核细胞、淋巴细胞计数,肺湿干重比值,血清、BALF中IL-5含量均明显升高,IFN-γ含量及AQP5表达水平均明显降低,差异均有统计学意义(P<0.01);与模型组比较,黄芪多糖中、高剂量组和地塞米松组小鼠BALF细胞总数,中性粒细胞、嗜酸性粒细胞、单核细胞、淋巴细胞计数,肺湿干重比值,血清、BALF中IL-5含量均明显降低,IFN-γ含量及AQP5表达水平均明显升高,差异均有统计学意义(P<0.01)。结论黄芪多糖可减轻支气管哮喘模型小鼠气道炎症,改善细胞因子的分泌,上调AQP5的表达,加快肺泡内液体的清除,减轻肺组织细胞受损程度,对肺组织具有保护作用。Objective To investigate the effects of astragalus polysaccharides( APS) on airway inflammation and expression of aquaporin 5( AQP5) in model mice with bronchial asthma. Methods 60 male C57 BL/6 J mice were randomly divided into 6 groups: blank group,model group,dexamethasone group,APS low-,medium-and highdose groups,10 cases in each. Except control group,mice in other groups were intervened with intraperitoneal injection of ovalbumin( OVA) together with aluminum hydroxide and OVA aerosol inhalation to establish bronchial asthma models. After modelling,the blank group and model group were given saline( 0.2 m L) by gavage; 3 dose groups were given APS at 100.0,200.0 and 300.0 mg/kg respectively; dexamethasone group was given intraperitoneal injection of dexamethasone sodium phosphate( 0. 8 mg/kg). After 30 days of corresponding drug intervention,total cell number,differntial count in bronchoalveolar lavage fluid( BALF) and lung wet-dry-weight ratio( W/D) of all groups were compared; ELISA was used to measure the content of IL-5,IFN-γ as well as the expression of AQP5 in serum and BALF of mice. Results Compared with blank group,total cell number,neutrophils,eosinophils,monocytes,lymphocytes counts,W/D,IL-5 of model group increased significantly,while IFN-γ,AQP5 significantly decreased( P0.01); compared with model group,total cell number,neutrophils,eosinophils,monocytes,lymphocytes counts,W/D,IL-5 of APS medium-and high-dose groups,dexamethasone group decreased obviously,while IFN-γ,AQP5 increased. The differences were all significant( P0.01). Conclusion APS can ease the airway inflammation,improve the secretion of cytokines and increase the expression of AQP5,accelerate the alveolar liquid clearance,alleviate the damage to cell of lung tissue and protect the lung tissue.
