体外受精-胚胎移植失败者子宫内膜整合素β_3及细胞间隙连接蛋白43的表达研究

摘　　要：目的探讨子宫内膜中整合素β_3、细胞间隙连接蛋白43(connexin 43,Cx43)表达水平与妊娠结局的关系,进而分析其与子宫内膜容受性的相关性。方法选择行体外受精-胚胎移植失败者35例作为观察组,既往有正常生育史者15例作为对照组,于黄体中期行宫腔镜检查并取内膜,应用免疫组化方法检测子宫内膜整合素β_3和Cx43表达的HSCORE评分并进行组间比较。结果观察组与对照组子宫内膜腔上皮整合素β_3表达的平均HSCORE评分分别为0.22±0.34和0.57±0.48,组间比较差异有显著性(P=0.01),而两组子宫内膜腺上皮的整合素β_3表达未见明显差别(P>0.05)。观察组与对照组子宫内膜间质细胞Cx43表达的平均HSCORE评分分别为2.11±0.83和1.49±0.77,组间比较差异有显著性(P=0.02),而两组子宫内膜上皮部的Cx43表达未见明显差别(P>0.05)。结论体外受精-胚胎移植者妊娠失败可能与子宫内膜腔上皮整合素β_3表达降低、子宫内膜间质细胞Cx43表达升高有关。Objective To evaluate the relationship between expression of endometrial integrin β3, connexin 43 and the outcome of pregnancy. Method 35 women with in vitro fertilization（IVF） failure were selected as study group, while 15 fertile women who underwent hysteroscopy were selected as control group. Immunohistochemical staining was performed for the expression of integrin β3 and connexin 43 in endometrial samples. Result The average HSCORE of integrin β3 in the endometrial luminal epithelium were 0.22±0.34 in the study group, and 0.57±0.48 in the control group.The difference was significant（P=0.01）. In the endometrial glandular epithelium, there is no difference in the expression of integrin β3 between the two groups. The average HSCORE of Cx43 in the endometrial stroma were 2.11±0.83 in the study group, and 1.49±0.77 in the control group.The difference was significant（P=0.02）. In the endometrial epithelium, there is no difference in the expression of Cx43 between the two groups. Conclusion The failure of IVF-ET. might be related with the low level of integrin β3 in the endometrial luminal epithelium and high level of Cx43 in the endometrial stroma.
