摘 要:目的:探讨滋阴/补阳方对多囊卵巢综合征(PCOS)大鼠体内促炎细胞因子的影响。方法:建立PCOS大鼠模型,分为模型组、滋阴方组、补阳方组及空白组共4组,检测血清性激素、血清及下丘脑、垂体和卵巢组织中IL-6、TNF-α的表达。结果:血清IL-6浓度模型组显著高于空白组(P<0.01),两中药组显著性低于模型组(P<0.01)。血清TNF-α浓度模型组显著性高于空白组(P<0.05),补阳方组显著性低于模型组(P<0.05)。下丘脑和卵巢组织中IL-6基因模型组表达显著性高于空白组(P<0.01),两中药组显著性低于模型组(P<0.01)。下丘脑组织中TNF-α基因模型组表达显著性高于空白组(P<0.01),滋阴方组显著性低于模型组(P<0.01)。卵巢组织中TNF-α基因模型组表达显著性高于空白组(P<0.05),补阳方组显著性低于模型组(P<0.05)。结论:滋阴/补阳方可以降低PCOS大鼠血清IL-6、TNF-α浓度,显著下调下丘脑和卵巢IL-6、TNF-α基因的表达丰度以缓解机体炎症状态,改善糖代谢和内分泌紊乱。Objective: To investigate the effect of Ziyin/Buyang Formula on proinflammatory cytokines in polycystic ovary syndrome(PCOS) rats. Methods: the PCOS rat model was established and divided into four groups: model control group(II), Ziyin prescription group(III), Buyang prescription group( IV) and blank control group( I). Expression of proinflammatory cytokines in serum sex hormones, serum and tissues was detected. Results: the concentration of serum IL-6 in group II was significantly higher than that in group I(P〈0.01), while that in groups III and IV was significantly lower than that in group II(P〈0.01). The serum TNF-α concentration in group II was significantly higher than that in group I(P〈0.05), while that in group IVwas significantly lower than that in group II(P〈0.05). The expression of IL-6 gene in group II was significantly higher than that in group I(P〈0.01), while that in groups III and IV was significantly lower than that in group II(P〈0.01). The expression of TNF-αgene in group II was significantly higher than that in group I(P〈0.01) and group III was significantly lower than that in group II(P〈0.01). The expression of TNF-α gene in group II was significantly higher than that in group I(P〈0.05), while that in group IV was significantly lower than that in group II(P〈0.01). Conclusion: Ziyin/Buyang Formula can reduce the concentrations of IL-6 and TNF-α in serum of PCOS rats, significantly lower the expression abundance of IL-6 and TNF-α genes in hypothalamus and ovary in order to relieve the inflammatory state of the body and improve glucose metabolism and endocrine disorders.
