摘 要:目的:观察归脾汤的抗抑郁作用,并探讨其抗抑郁的作用机制。方法:健康雄性SD大鼠60只,进行糖水适应,建立糖水偏好基线,随机分为对照组和模型组,对照组常规饲养,模型组采用慢性不可预见性温和应激方法结合孤养3周建立抑郁模型。模型建立后,将模型组大鼠随机分为模型组、氟西汀组、归脾汤组,分别给予0.9%氯化钠溶液、氟西汀、归脾汤灌胃。给药4周后,对各组大鼠进行糖水偏好实验、强迫游泳实验、旷场实验、新奇摄食抑制实验等行为学检测,并观察各组大鼠海马CA3区脑源性神经营养因子(BDNF)水平的表达。结果:归脾汤组糖水偏好较模型组明显增高(ANOVA,F=12.55,P<0.01)。新奇摄食抑制实验中,归脾汤组的摄食时间与对照组相近,较模型组明显缩短(ANOVA,F=3.98,P=0.02);旷场实验中,归脾汤组大鼠中央区穿越次数较模型组明显增多(ANOVA,F=4.53,P=0.01)。归脾汤组的BDNF蛋白和细胞表达较模型组显著增多。结论:归脾汤可能通过增加抑郁大鼠海马CA3区BDNF水平发挥抗抑郁作用。Objective: To observe the antidepressant function and explore the possible mechanisms. Methods: Sixty healthy male SD rats were given sucrose experiment and establish a baseline of sucrose preference, and then they were randomly divided into normal control group and model group. The normal control group was normally raised, and the model group adopted the chronic unpredictable mild stress method combined with the social isolation 3 weeks to establish the depression model. After the model was established, they were randomly divided into model group(CUMS group), fluoxetine(FLU) and Guipi Decoction(GPT-H group), and then were intragastric administration physiological saline, fluoxetine and Guipi Decoction respectively. After four weeks, each group was given sucrose preference test, forced swimming test, open field test and novel feeding inhibition test for behavior test, and the expression of BDNF level in hippocampus of rats was determined. Results: In sucrose preference test, Guipi Decoction group was significantly higher than that in the model group(ANOVA, F=12.55, P〈0.01), and there was no significant difference with the control group. In novel feeding inhibition test, the feeding time of Guipi Decoction group(131.14) was similar to the control group, and it was significantly shorter than the CUMS group(ANOVA, F=3.98, P=0.02). In open field test, the number of crossings in central region of Guipi Decoction(10.10) was significantly increased than the CUMS group(ANOVA, F=4.53, P=0.01). There was no significant difference in food consumption in 5 minutes(F=0.91, P=0.45). The expression of BDNF protein and cells of Guipi Decoction group were significantly increased than the CUMS group, and there was no significant difference with control group(ANOVA, F=8.13, P〈0.01). Conclusion: Guipi Decoction may play an anti-depressant role by increasing the BDNF level in the hippocampus CA3 area of depressed rats.
