摘 要:目的:探讨安神定志灵方对自发性高血压大鼠(SHR)前额叶、纹状体及海马中Akt/GSK-3β/β-catenin信号通路的影响。方法:将50只SHR大鼠随机分为模型组、利他林组(2mg/kg)、安神定志灵低、中、高剂量组(6.7、13.4、26.7g/kg),每组10只,另将10只Wistar京都大鼠设为正常组。每日灌胃2次,治疗4周后取大鼠脑组织。分别用蛋白质印迹法(Western Blot)、免疫组织化学法及实时定量荧光PCR检测各组大鼠前额叶、纹状体、海马组织中Akt、GSK-3β和β-catenin的表达水平。结果:与正常组比较,模型组大鼠前额叶、纹状体中β-catenin、Akt的蛋白及mRNA表达降低,GSK-3β相关表达增加;海马区β-catenin、Akt的OD值显著性降低(P<0.05),GSK-3β的OD值显著性升高(P<0.05)。治疗后与模型组比较,利他林及安神定志灵组能够增加大鼠前额叶、纹状体中β-catenin、Akt的蛋白及mRNA表达,降低GSK-3β的相关表达;增加大鼠海马区β-catenin、Akt的OD值,降低GSK-3β的OD值。结论:安神定志灵可以影响Akt、GSK-3β、β-catenin表达水平,其药效可能与抑制细胞凋亡、保护神经元存在关联。Objective: To explore the effects of Anshen Dingzhi Ling(ADL) Decoction on Akt/GSK-3β/β-catenin signaling pathway of the prefrontal, striatum, and hippocampus in spontaneously hypertensive rats(SHR). Methods: Fi Fty SHR were randomly divided into model group, ritalin group(2 mg/kg), Anshen Dingzhi Ling Decoction low, medium, high dose group(6.7, 13.4, 26.7 g/kg respectively), with ten rats in each group, and ten Wistar-Kyoto rats were set as the normal group. The rats were treated 2 times a day for four weeks, and then they were sacrificed to take brain tissues. Western Blot, immunohistochemistry and real-time PCR were used to detect the expression levels of Akt, GSK-3β,β-catenin in SHR striatum, prefrontal cortex and hippocampus. Results: Compared with the normal group, the protein and mRNA expression of dex and Akt in the frontal lobe and striatum of the model group were decreased, and GSK-3 related expression was increased. In the hippocampal area, the OD value of caec-catenin and Akt was significantly decreased(P〈0.05), while the OD value of GSK-3 was significantly increased(P〈0.05). After treatment, compared with the model group, ritalin and anshen dingling group can increase the protein and mRNA expression of-catenin and Akt in the frontal lobe and striatum of rats, and reduce the related expression of GSK-3. Increase the OD value of the hippocampal region of the rat and decrease the OD value of GSK-3. Conclusion: Anshen Dingzhi Ling Decoction can influence the expression levels of Akt, GSK-3β and β-catenin, and its efficacy may be associated with inhibiting apoptosis and protecting neurons.
