摘  要:目的观察富血小板血浆(PRP)在新生乳鼠皮肤组织及毛囊发育过程中的影响。方法取新生C57BL/6乳鼠40只,随机分为PRP实验组和生理盐水对照组,每组20只。在背部正中部位区域,实验组0.05ml血小板浓度为1×10^9/mlPRP单点皮内注射,对照组等量注射生理盐水。分别于第3、5、7、10天时每组各取5只动物进行大体观察,并切取全层皮肤组织石蜡包埋常规苏木素-伊红(HE)、Masson染色,对组织切片进行组织学观察拍摄,使用Image J软件测量皮肤灰度值,ipp软件对真皮及表皮的厚度进行测量。结果在第3、5、7天时大体观察PRP组小鼠皮肤发育颜色的灰度值(9723.30±874.87、13371.25±1511.42、16794.70±1899.03,均P=0.000),其现象改变较对照组更早发生,其皮肤颜色灰度值检测统计结果与大体观察相符,同时,第10天时,大体观察发现PRP组小鼠毛发的密度也明显高于对照组。HE染色结果显示PRP组的小鼠3、5、7、10d皮肤组织表皮厚度(0.435±0.011、0.460±0.009、0.497±0.008、0.052±0.014,P=0.005、0.000、0.000、0.016),均高于对照组。PRP组的小鼠3、5、7、10d皮肤组织真皮厚度(0.744±0.011、1.168±0.016、1.820±0.011、1.717±0.012,P=0.000、0.000、0.000、0.034),在同天数的发育过程中厚度明显高于对照组。PRP实验组3、5、7、10d毛囊数量(18.33±1.77、25.33±1.02、34.33±1.58、39.33±1.23,P=0.000、0.000、0.005、0.007),与对照组比较,毛囊数量明显增加。并且实验组皮肤组织内毛囊在同天数发育程度优于对照组。Masson染色结果显示早期皮肤组织发育过程中,PRP组内新生胶原的分泌量明显高于对照组,并且其胶原的排列更早趋向于成熟皮肤组织内胶原的排列状态。结论PRP有助于皮肤及毛囊的发育与成熟。Objective To observe the effect of platelet- rich plasma (PRP) on the skin tissue and follicle development of neonatal mice. Methods Forty neonatal C57BL/6 mice were randomly divided into PRP experimental group and normal saline control group, 20 mice in each group. In the middle of the back, single - point intradermal injection of 0. 05 ml PRP with blood platelet counts of 109/ml was carried out in the experimental group, and equivalent injection of normal saline was carried out in the control group. The general observation was done on 5 animals each at the 3rd, 5th, 7th and 10th day, and the full thickness skin tissue was cut for paraffin embedding and conventional hematoxylin and eosin (HE) and Masson staining. The histological changes of the tissue sections were observed and photoed. Image J software was used to measure the gray level of skin, and ipp software to measure the thickness of the dermis and the epidermis. Results At the 3rd day, 5th day, and 7th day, the gray values (9 723.30 ± 874.87, 13 371.25 ± 1 511.42, 16 794. 70 ± 1 899.03, mean P = 0. 000) of the skin development color of the mice in PRP group were generally observed. The phenomenon of the changes in the experimental group oecurred earlier than that in the control group. The statistical results of the grey values of the skin color were in accordance with the general observation. Simultaneously, at the 10th day, the general observation revealed that the hair density value of the mice in PRP group was significantly higher than that of the mice in the control group. The HE staining results showed that at the 3rd day, 5th day, 7th day, and 10th day, the values of skin tissue epidermal thickness (0. 435 ± 0. 011, 0. 460 ± 0. 009, 0. 497 ± 0. 008, 0. 052 ± 0. 014 ; P = 0. 005, 0. 000, 0. 000, 0. 016 ) of the mice in the experimental group were all higher than those of the mice in the control group. At the 3rd day, 5th day, 7th day, and 10th day, the values of the skin tissue dermis thickness (0. 744 ± 0. 011, 1. 168 ± 0
