摘 要:目的:本文旨在探索四种不同提取方法对武当山地区3种娑罗子(野生、半野生、人工种植)中的β-七叶皂苷A、B的提取效果,并同时分析两种化合物的含量。方法:分别采用醇提法、回流提取法、超声提取法、索氏提取法等方法提取娑罗子中的β-七叶皂苷A、B,采用高效液相色谱法测定各种提取方法所得β-七叶皂苷A、B的含量。色谱条件:Sun Fire TM LC-C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%磷酸水溶液(37∶63);检测波长为UV 220 nm,流速为1.0 mL/min,柱温为35℃。结果:不同提取方法对娑罗子中β-七叶皂苷的提取率影响较大,其中索氏提取法提取效率最高,而超声提取法提取效率最低。结论:采用不同的提取方法对含量测定的结果影响较大,索氏提取法为本次试验的最佳提取方法。Objective To explore four different extraction methods of β-aescin A,B from three different varieties of Semen Aesculi in Wudang Mountain area( wild,semi-wild,artificial planting) and determine the content of β-aescin A,B,and to determine the contents of β-aescin A and β-aescin B,simultaneously. Methods The alcohol extraction method,reflux extraction method,ultrasonic extraction method and soxhlet extraction method were used respectively. The content of β-aescin was determinated by HPLC method. Chromatographic conditions: a Sun Fire TM LC-C18 column( 4.6 mm×250 mm,5μm) was used. The solution of acetonitrile-0.2% phosphoric acid solution( 37 ∶ 63) was used as the mobile phase at a flow rate of 1.0 mL/min and the detection wavelength was 220 nm. Separation was performed at 35 ℃. Results The efficiency of different extraction methods on the extraction yield of β-aescin were different. The efficiency of soxhlet extraction method was high and the ultrasonic extraction method was low. Conclusion The extraction method plays an important role in the extraction of β-aescin from Semen Aesculi,in which the soxhlet extraction method was the best.
