摘 要:AP2/EREBP类转录因子为植物所特有的一类转录因子,参与了植物的发育和胁迫途径。本研究利用枣的全基因组数据库,通过生物信息学手段鉴定枣的AP2/EREBP家族的全部成员,并分析AP2/EREBP转录因子家族保守域特点与功能及表达情况,为枣的遗传改良抗性育种提供理论依据。利用在线软件EBI、TAIR、Hmmer3.0、SMART、Pfam、MEME、ProtParam、SOPMA、SignalP4.1Sever和String网站,以及ClustalX2、BioEdit、MEGA6.0、MapInspect软件,对枣的AP2/EREBP转录因子家族的类型、结构域、系谱进化、序列元件、蛋白理化性质及二级结构、信号肽分析、基因定位、基因芯片表达和蛋白功能联系预测进行了分析。分析表明,枣全基因组中有127条AP2/EREBP转录因子,根据其结构域划分为4个亚家族:AP2、RAV、DREB及ERF,各家族成员数分别为:17、6、50及54。进行多序列比对和系谱进化分析,发现2个亚家族各分为6个亚族。生物信息学分析表明,AP2/EREBP家族蛋白均为亲水性蛋白、非分泌型蛋白,且该家族蛋白富含酸性氨基酸;基因定位表明,该家族基因在12条染色体上呈不均匀分布,有染色存在串联复制现象;对127个枣AP2/EREBP蛋白之间的功能联系网络进行了系统预测,分析预测了相关基因的功能和多个基因间的联系;基因芯片表达表明,AP2/EREBP转录因子在低温、高温、光照、遮阴、紫外线、UV敏感处理及外源ABA处理条件下均能被诱导表达,且在根、叶、花和果4个器官中均有显著性表达。枣AP2/EREBP基因家族结构高度保守,对外界胁迫有显著性表达,可能在枣的生长发育及其在枣的多种生理反应信号转导中发挥着重要作用,且各家族成员之间在植物生长发育过程和胁迫响应及激素信号转导途径具有交叉作用。AP2/EREBP transcription factor is a kind of transcription factor peculiar to plants and participates in plant development and stress. In this research, all members of AP2/EREBPfamily were identified by bioinformatics analysis in terms of the whole genome data of Ziziphus jujube.The characteristics, function and expression of AP2/EREBP transcription factor family conserved domain were analyzed, which provided the theoretical basis for genetic improvement and resistance breeding of Ziziphus jujube. The types, structural domain, genealogical evolution, sequence elements, protein physicochemical properties and secondary structure, signal peptide analysis, gene localization, gene expression and protein functional contact prediction of Ziziphus jujubeAP2/EREBP transcription and factor family were analyzed by EBI, TAIR, Hmmer3.0, SMART, Pfam, MEME, ProtParam, SOPMA, SignalP4.1 Sever and String web, as well as ClustalX2, BioEdit, MEGA6.0 and MapInspect software. The results showed that there were 127 AP2/EREBP transcription factors in the whole genome of Ziziphus jujube, which were divided into 4 subfamilies: AP2, RAV, DREB and ERF according to their structural domains. The members of each subfamily were 17, 6, 50 and 54, respectively. The analysis of multiple sequence alignment and genealogical evolution showed that 2 subfamilies were divided into 6 subfamilies. Bioinformaties analysis showed that all AP2/EREBP family proteins were hydrophilic protein, non-secreted protein, and the family proteinswere rich in acidic amino acids; Gene localization showed that the family genes were unevenly distributed on 12 chromosomes, and some genes had tandem duplication; The functional connection network of 127 Ziziphus jujubeAP2/EREBP proteins was systematically predicted, as well as the function of related genes and the connection among several genes were analyzed; gene expression showed that AP2/EREBP transcription factors could be induced for expression under the conditions of low temperature, high temperature, light, shade, ul
