摘 要:通过对蒙古扁桃16个居群的叶绿体DNA(cpDNA)的trnH-psbA及nrDNAITS序列进行测序,对两种序列进化速率、核苷酸多样性及单倍型系统发育关系进行了初步研究。结果表明:蒙古扁桃cpDNAtrnH-psbA序列总长度为306bp,共得到52处变异位点,变异率达到17.05%,共检测到11种单倍型;nrDNAITS序列总长度为662bp,共得到6处变异位点,变异率达为0.91%,共检测到9种单倍型。经比较发现,蒙古扁桃nrDNAITS序列较cpDNAtrnH-psbA序列保守,变异速率较慢。cpDNAtrnH-psbA及nrDNAITS序列单倍型系统发育关系的各支系的支持率均较高。According to sequencing trnH-psbA and nrDNA ITS sequences of 16 populations of chloroplast DNA (cpDNA) in Prunus mongolica, the study of evolutionary rates, nucleotide diversity and phylogenetic relationship of haplotypes were carried out. Results showed that the total length of cpDNA trnH-psbA sequence of P. mongolica was 306 bp, which had 52 variable sites with a mutation rate of 17.05%, and had a total of 11 haplotypes; the total length of nrDNA ITS sequence of P. mongolica was 662 bp, which had 6 variable sites with a mutation rate of 0.91%, and had a total of 9 haplotypes. The nrDNA ITS sequence of P. mongolica was more conserved and evolved slower than the cpDNA trnH-psbA sequence through the comparative analysis. The approval rating for each branch of haplotype phylogenetic relationship of cpDNA trnH-psbA and nrDNA ITS was high.
