摘 要:目的:采用UPLC指纹图谱结合多指标综合评分法优选肝乐颗粒的醇提工艺。方法:采用正交设计法,以绿原酸和芍药苷的含量、干膏得率、正交样品指纹图谱中9个主要共有峰的总面积为指标,考察溶剂倍数、乙醇体积分数、提取时间和提取次数四因素对肝乐颗粒醇提工艺的影响。采用综合评分法进行数据分析,建立9个正交样品的指纹图谱,并对其用中国药典委员会颁布的中药色谱指纹图谱相似度评价系统软件(版本:2004A)和SPSS 19.0软件分别进行相似度分析、聚类分析,优选肝乐颗粒的最佳醇提工艺。结果:肝乐颗粒的最佳醇提工艺为:溶剂倍数为10倍量的60%乙醇回流提取3次,每次2 h;9个正交样品的相似度均大于0.97;聚类分析表明正交样品可大致聚成4类。结论:优选的提取工艺合理、可行,有效成分含量高,建立的指纹图谱重现性好,验证了优选醇提工艺的可行性,为肝乐颗粒醇提工艺的确定提供实验依据。Objective:To optimaize the alcohol extraction technique for Ganle Granule and confirm the optimum extraction process parameters by develop an UPLC fingerprint combined with multi index comprehensive weighted score evaluation. Methods:The orthogonal test was employed taking the contents of chlorogenic acid,paeoniflorin,dry extract rate and the total area of the 9 main peaks in the fingerprints of orthogonal samples as the indexes and the influences on extraction results of soaking time,solvent dosage and extraction times were investigated,and were analyzed by multi-index comprehensive scoring method. The fingerprint of the 9 orthogonal samples was established,and the chinese pharmacopoeia commission promulgated the chromatographic fingerprint similarity evaluation system software(version:2004 A)analyzed the similarity analysis,SPSS 19.0 software analyzed cluster analysis. Results:The optimal extracting process was selected as follows:10 times of alcohol consumption,60% ethanol reflux extraction three for three times,and two hour each time,the similarity of the 9 orthogonal samples was more than 0.97;the cluster analysis showed that the orthogonal samples can be roughly clustered into 4 categories. Conclution:The optimized extraction process is reasonable and feasible,and the effective component content is high,the fingerprint reproducibility is good,which verifies the feasibility of optimizing the alcohol extraction process,and provides the experimental basis for the determination of ethanol extraction process of the Ganle Granule.
