摘  要:种子休眠期长是导致珙桐自然繁殖力低下的重要原因之一。珙桐内果皮在发育后期快速木质化形成坚硬而致密的结构,可能是决定种子休眠时间的重要因素,而这一过程的调控机制尚不明确。本研究小组前期通过对珙桐内果皮发育的4个时期进行转录组测序,发现CCo AOMT基因家族与内果皮快速木质化关系密切。本研究从转录组中筛选到11个珙桐CCo AOMT基因家族基因,并利用生物信息学方法对筛选到的基因进行表达模式、序列特征、细胞定位和系统进化分析。分析显示,11个珙桐CCo AOMT基因根据表达模式可分为2种类型,筛选其中6个表达差异显著的CCo AOMT基因进一步分析其表达规律,最后确定其中差异表达最显著的Cluster-149.63013序列进行克隆及生物信息学分析。结果表明,该基因开放阅读框全长为744 bp,编码247个氨基酸,其氨基酸序列含有植物甲基化酶所共有的A、B、C基序及CCo AOMT蛋白特有的标志性序列D、E、F、G和H,其氨基酸序列与辣椒的CCo AOMT蛋白同源性最高,将其命名为Di CCo AOMT1。本研究克隆得到的Di CCo AOMT1基因可能是珙桐内果皮木质素快速积累的关键调控基因。The long period of seed dormancy is one of the most important reasons leading to the low fertility of dove tree(Davidia involucrata). The hard and dense endocarp formed in the late developmental state of Davidia fruit might be an important factor to determine the long period of seed dormancy, while the regulation mechanism of this process is unclear. According to the transcriptome data of Davidia endocarp previously obtained by our group, the CCo AOMT gene family was found closely related to the rapid lignification of endocarp. Eleven CCo AOMT genes were identified from the transcriptome data. The expression patterns of these genes in different developmental stages of endocarps were analyzed. The results showed that there are 2 types of CCo AOMT genes in Davidia.Based on the expression patterns, 6 differentially expressed CCo AOMT genes were selected, and Cluster-149.63013, the tanscript with most significant expression difference, was cloned. The sequence of Cluster-149.63013 contains a 744 bp open reading frame and encodes a 239-amino acid protein. The amino acid sequence of Di CCo AOMT1 contains A, B and C motifs, which are common in plant methyltranferase, and the unique landmark motifs D, E, F, G and H of the CCo AOMT family. Phylogenetic analysis showed that its amino acid sequence has the highest homology with that of Capsicum annuum. Therefore, it was named as Di CCo AOMT1. The identified Di CCo AOMT1 in this study may be a key regulator involved in the rapid lignin accumulation in Davidia endocarps.
