摘  要:目的探讨红景天苷对力竭大鼠心肌收缩力的影响,为临床力竭心脏损伤防护提供新的思路。方法40只雄性SD大鼠随机分为4组(n=10):对照组,力竭组,红景天苷A、B组。Thomas方法制备力竭大鼠模型。红景天苷A、B组分别给予15、30mg/(kg·d)红景天苷溶液,对照组、力竭组给予等体积0.9%NaCl溶液,腹腔注射2周。ELISA法检测各组血清肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、肌钙蛋白I(TnI)含量的变化。采用Langendorff装置分离大鼠心室肌细胞,以Ion Optix细胞收缩和钙离子浓度同步测定系统检测大鼠心肌细胞的收缩功能和钙瞬变,激光共聚焦显微镜线扫描成像技术记录心肌细胞自发性钙火花及肌质网钙含量。结果与对照组比较,力竭组大鼠心肌细胞钙瞬变、最大收缩幅度分别降低41%和45%(P<0.01),钙火花发生率增加了1.5倍(P<0.01),心肌细胞肌质网钙离子含量明显减少(P<0.01)。与力竭组比较,红景天苷A、B组大鼠心肌细胞钙瞬变分别增加了20%、38%,最大收缩幅度分别增加了25%、54%(P<0.01),钙火花发生率明显降低(P<0.01),心肌细胞肌质网钙离子含量明显增加(P<0.01);且红景天苷B组各指标均明显优于红景天苷A组(P<0.05或P<0.01)。结论红景天苷可改善力竭运动造成的心肌细胞收缩力减弱,其机制与细胞内钙离子调控有关。Objective To investigate the effect of salidroside on myocardial contractility in exhausted rats for providing a new idea on protection of cardiac exhaustive injury. Methods Forty male Sprague-Dawley rats were randomly divided into 4 groups(n=10): control group, exhaustive group, salidroside A and B group. Thomas method was employed to establish the exhaustive animal model. Salidroside A and B groups were given salidroside solution 15 mg/(kg·d) and 30 mg/(kg·d) for 2 weeks, respectively, and the same volume of 0.9% NaCl was given to the exhaustive and control groups by intraperitoneal injection for 2 weeks. Serum creatine kinase(CK), creatine kinase isoenzyme(CK-MB) and troponin I(TnI) levels were detected by ELISA. Langendorff apparatus was used to separate the rats' ventricular myocytes. Rats' myocardial cell contractile function and calcium transient were detected with Ion Optix cell contraction and calcium concentration synchronous measurement system. Laser confocal microscopy line scanning imaging was performed to record the cardiac myocytes spontaneous calcium spark and sarcoplasmic reticulum calcium content. Results Compared with the control group, the calcium transients and maximal contractions amplitude of myocardial cells decreased by 41% and 45%(P〈0.01), the incidence of calcium sparks increased by 1.5 times(P〈0.01), the sarcoplasmic reticulum calcium content decreased significantly(P〈0.01) in rats of exhaustive group. Compared with the exhaustive group, the calcium transients in myocardial cells increased by 20% and 38%, respectively, and the maximal contraction amplitude increased by 25% and 54%(P〈0.01), the incidence of calcium sparks was significantly higher(P〈0.01), and the sarcoplasmic reticulum calcium content increased significantly(P〈0.01) in rats of salidroside A and B groups.
