摘  要:目的探讨重度抑郁症患者circRNA表达水平及其与抑郁症严重程度的关系。方法选取抑郁症患者(抑郁症组,n=5)和健康人(n=5)进行基因芯片筛查,确定10种存在差异表达的circRNA。对抑郁症患者行进一步PCR验证,并采用汉密尔顿抑郁量表(HAMD)对其抑郁症状进行评估。结果 circRNA_103636的ΔCt值与焦虑/躯体化、认知障碍呈明显负相关(P<0.05)。circRNA_100679的ΔCt值与体重、绝望感呈明显正相关(P<0.05)。circRNA_100679对抑郁症组体重、绝望感有显著预测作用,对体重、绝望感变异的解释率分别为13.9%、11.1%(P<0.05或P<0.01)。circRNA_104121、circRNA_102802和circRNA_104953对迟滞有显著预测作用(P<0.05),可解释迟滞因子变异的16.9%。circRNA_104600、circRNA_103636对睡眠障碍有显著预测作用(P<0.05),可解释睡眠障碍变异的15.4%。抑郁症高分组与低分组的circRNA_002143、circRNA_102802、circRNA_103636、circRNA_100679和circRNA_104953表达水平不同,差异有统计学意义(P<0.05)。ROC分析表明,抑郁症患者外周血circRNA_100679表达水平对其症状严重程度有中等预测效力(AUC=0.708,P<0.05,95%CI:0.523~0.894)。结论 circRNA_100679表达水平对抑郁症严重程度有较好的预测价值。hsa_circRNA_002143、circRNA_103636表达水平与患者的抑郁症状有密切关系。Objective To explore the correlation of circRNA expression level and severity of depression in major depressive disorder(MDD) patients in order to provide solid scientific evidences for diagnosis and prognosis evaluation of MDD. MethodscircRNA array was performed in peripheral blood mononuclear cells(PBMCs) obtained from 5 MDD patients and 5 controls, and then real-time fluorescence quantitative reverse transcription polymerase chain reaction(q RT-PCR) was used to verify the top 10 circRNAs with the highest fold change values. All the MDD patients were assessed by Hamilton Depression Scale(HAMD). Results The change of expression level of circRNA103636 was negatively correlated to anxiety/somatization and cognitive disorder; the change of expression level of circRNA100679 showed a significantly positive correlation with weight and hopelessness(P〈0.05). Regression analysis demonstrated that the alteration of circRNA100679 expression accounted for 13.9% and 11.1% respectively of weight and hopelessness; expression levels of circRNA104121, circRNA102802 and circRNA104953 could predict retardation(P〈0.05) and accounted for 16.9% of variances; expression levels of circRNA104600 and circRNA103636 could significantly predict variance of sleep disorder(P〈0.05, the accounting percentage was 15.4%). Expression levels of circRNA002143, circRNA102802, circRNA103636, circRNA100679 and circRNA104953 showed statistical difference between upper-score group and lowerscore group(P〈0.05). ROC analysis suggested expression level of circRNA100679 in PBMCs had neutral predictive efficacy on depressive severity(AUC=0.708, P〈0.05, 95% confidence inter val: 0.523-0.894). Conclusions Altered expression of circRNA100679 may be involved in MDD pathogenesis and may serve as a biomarker for MDD. Moreover, expression levels of hsacircRNA002143 and circRNA103636 might be closely related to severity of symptoms in MDD patients.
