摘  要:针对采自全国7个自然保护区共计11个野生金针菇和2个常见栽培金针菇菌株,进行ITS-PCR和产物测序,经过与Gen Bank上已知序列的比对,结合传统形态特征鉴定,确定各菌株的拉丁名,并用MEGA6.0进行ITS序列的遗传距离分析和系统发育树构建。结果表明,金针菇的ITS1比ITS2进化速率快,变异位点和简约信息位点差异较大;13株金针菇种内遗传距离为0~0.014,种间遗传距离为0.029~0.045;ITS系统发育树也支持将13个菌株分为3个类群,即3个种,这与分子鉴定和传统形态鉴定结果相吻合,且同一菌种地域分布越近的菌株越容易聚为一支。研究表明,ITS序列分析可用于野生金针菇的分子鉴定和种间遗传多样性分析。In order to calculate the genetic distance between strains of Flammulina sp. and phylogenetie relationship of these strains, ITS sequences of eleven wild Flammulina sp. collected from the National Nature Preserve and two common cultivated Flammulina velutipes were amplified and sequenced. Genetic distance and phylogenetic relationship were analyzed by MEGA6.0. The results showed that the evolution rate of ITS1 is faster than ITS2, and the variable sites and parsimony informative sites of ITS1 differs greatly from ITS2. Intraspecific genetic distance of 13 experimental strains was 0-0.014, and the interspecific genetic distance of them was 0.029-0.045. Phylogenetic tree based on ITS sequences supported the view that 13 experimental strains interspecific could be divided into three broad categories, the closer of strains in regional relationship was also closer in phylogenetic relationship. The research indicated that ITS sequence analysis could be used for the molecular identification and genetic diversity analysis of wild Flammulina sp.
