摘  要:目的探究拉莫三嗪抗抑郁作用及其机制。方法分别向抑郁模型小鼠腹腔注射拉莫三嗪(7,15,30ml/kg)以及氟西汀(20mg/kg),通过强迫游泳试验(FST)、悬尾实验(TST)以及旷场实验(OFT)观察拉莫三嗪的抗抑郁作用;预先向抑郁模型小鼠腹腔注射N-甲基-D-天冬氨酸(NMDA)及NMDA受体拮抗剂MK-801(MK)、Mg SO4、氯胺酮再给予拉莫三嗪,探究拉莫三嗪抗抑郁作用的机制。结果拉莫三嗪具有抗抑郁作用,且呈量效依赖关系;NMDA能够抑制拉莫三嗪抗抑郁的作用;MK-801、氯胺酮、Mg SO4能够促进拉莫三嗪抗抑郁的作用。结论拉莫三嗪可能是通过抑制NMDA受体作用发挥抗抑郁作用。Objective To investigate the mechanism of anti-depression of lamotrigine and its mechanism. Methods Lamotrigine at 7,15,30 mg/kg and fluoxetine at 20 mg/kg were injected intraperitoneally into the mice. The antidepressant effect of lamotrigine was observed by the methods of forced swimming test( FST),tail suspension test( TST)and open field test( OFT). The mice pretreated with MK,NMDA,MgSO4 and ketamine were given lamotrigine to explore the mechanism of Lamotrigine as an antidepressant. Results Lamotrigine had antidepressant effect which was dosedependent.MK-801,ketamine and MgSO4 were able to promote the antidepressant effect of lamotrigine,but NMDA were able to inhibit the antidepressant effect of lamotrigine. Conclusion The antidepressant effect of lamotrigine may be induced by inhibiting of NMDA receptor.
