摘  要:目的:探究N-甲基-D-天冬氨酸受体(NMDAR)蛋白含量及磷酸化水平在升降散治疗大鼠神经病理性疼痛过程中的作用机制。方法:构建大鼠右侧改良坐骨神经慢性缩窄性损伤(iCCI)经典动物模型,并采用不同剂量的升降散给iCCI模型大鼠进行灌胃处理,在给药第1、7、21天分别取脊髓组织,对NMDAR和pNMDAR水平进行检测。结果:不同时间点,正常组NMDAR及p-NMDAR相对含量较稳定,模型组NMDAR及p-NMDAR呈上升趋势,治疗组,尤其是高剂量组,呈现下降趋势,且在不同时间点,各组NMDAR及p-NMDAR相对含量存在显著性P〈0.05差异。结论:NMDAR及p-NMDAR参与了升降散治疗大鼠神经病理性疼痛过程中,通过抑制NMDAR水平及NMDAR磷酸化,有效的治疗了大鼠神经病理性疼痛。Objective: To study the role of NMDA receptor contents and phosphorylation in the mechanism of Shengjiang Powder treating neuropathic pain of rats. Methods: We set up chronic constriction injury of the sciatic nerve model,and used different doses of Shengjiang Powder to treat the iCCI model rats. On 1 d,7 d,21 d respectively,we tested the levels of NMDAR and p-NMDAR in spinal cord tissue. Results: At different time points,the normal group's NMDAR and p-NMDAR relative contents remained unchanged and the model group's NMDAR and p-NMDAR were on the rise. The treatment groups,especially the high dose group,showed a trend of decline. And at different time points,each group's NMDAR and p-NMDAR relative contents were significant( P 〈 0. 05). Conclusion: NMDAR and p-NMDAR participated in the mechansim of Shengjiang Powder treating neuropathic pain of rats,by inhibiting the NMDAR level and NMDAR phosphorylation. It is effective treatment for neuropathic pain of rats.
