从炎症因子角度研究定悸方对异丙肾上腺素诱导大鼠的作用机制

摘　　要：目的：从炎症因子角度出发,研究定悸方治疗异丙肾上腺素诱导左心室重构大鼠的作用机制。方法：采用异丙肾上腺素诱导建立左心室重构大鼠,分别给予蒸馏水（模型组）、中药定悸方（中药组）、卡托普利（西药组）、定悸方联合卡托普利（中西药组）灌胃治疗28 d后,ELISA法检测各组大鼠血清CRP、IL-1、IL-6、TNF-α水平,免疫组化法检测TGF-β1水平,HE染色观察左心室心肌形态学改变。结果：与模型组比较,中药组大鼠血清CRP、IL-1、IL-6、TNF-α表达水平降低,且中西药组效果最佳（P〈0.05）。定悸方能有效改善心肌TGF-β1表达水平,中西药组效果最佳（P〈0.05）,而中、西药组间无显著差异。结论：定悸方能通过降低炎症因子CRP、IL-1、IL-6、TNF-α、TGF-β1的水平来改善左心室肥厚程度,抑制心肌纤维化进程。Objective： From the point of view of inflammatory factors,to study the mechanism of Dingji Decoction in the treatment of isoproterenol-induced left ventricular remodeling in rats. Methods： The left ventricular remodeling rats were induced by isoproterenol and they were given distilled water（Model group）,Dingji Decoction group（Chinese medicine group）,Captopril group（Western medicine group）,Dingji Decoction combined with Captopril group（Chinese and Western medicine group） by gavage for 28 days. The levels of serum CRP,IL-1,IL-6 and TNF-α were measured by ELISA and the level of TGF-β1 was detected by immunohistochemistry. The morphological changes of left ventricular myocardium were observed by HE staining. Results： Compared with the model group,the levels of serum CRP,IL-1,IL-6 and TNF-α were decreased in the Chinese medicine group,and the combination of Chinese and Western medicine was the best（P〈0. 05）. Dingji Decoction can effectively improve the expression of TGF-β1 in myocardium,and the effect of Dingji Decoction combined with Captopril group was the best（P〈0. 05）,but there was no significant difference between Chinese medicine and Western medicine group. Conclusion： Dingji Decoction can effectively reduce the levels of inflammatory factors such as CRP,IL-1,IL-6,TNF-α and TGF-β1,improve the degree of left ventricular hypertrophy and inhibit the process of myocardial fibrosis.
