摘  要:目的探讨5-氨基酮戊酸光动力疗法(5-aminolevulinicacid photodynamic therapy,5-ALA-PDT)对RAW264.7细胞吞噬活性和活性氧自由基(reactive oxygen species,ROS)的影响。方法细胞实验研究用灭活痤疮丙酸杆菌诱导小鼠巨噬细胞株RAW264.7细胞,产生炎症细胞模型,给予不同浓度的5-ALA孵育RAW264.7细胞,并给予16 J/cm^2红光照射。采用中性红法检测巨噬细胞的吞噬活性,荧光显微镜观察细胞内活性氧离子的荧光强度,2',7'-二氯荧光黄双乙酸盐法(dihydrochloride acetyl acid dichloride,DCFH-DA)检测活性氧自由基水平。结果模型组吞噬活性比空白组高,随着5-ALA浓度升高,吞噬活性降低,呈剂量依赖性。荧光显微镜下5-ALA 0.12 mmol/L组细胞内有大量红色荧光。随着5-ALA浓度升高,ROS水平升高,呈剂量依赖性。结论 5-ALA-PDT可对RAW264.7细胞吞噬活性和活性氧自由基产生影响,可能影响5-ALA-PDT的疗效。Objective To explore the effect of 5-ALA-PDT on capability of phagocytosis and the level of ROS of murine macrophage RAW264.7 cells. Methods Murine macrophage RAW264.7 cells were induced by inactivated Propionibacterium acnes fluid to establish an inflammatory cell model, then the RAW264.7 cells were treated with different concentrations of 5-ALA combined with infrared radiation with a light dose of 16 J/cm^2. The capability of phagocytosis of RAW264.7 cells was detected by using neutral red method. Fluorescence intensity of cellular reactive oxygen species(ROS) was detected by fluorescence microscopy, the level of reactive oxygen species(ROS) was detected by dihydrochloride acetyl acid dichloride. Results The capability of phagocytosis of the model group was higher than that of the blank group. With the increasing concentration of 5-ALA, the capability of phagocytosis decreased in a dose-dependent manner. The result of fluorescence showed that the cells in the 0.12 mmol/L 5-ALA group had strong red fluorescence. With the increasing concentration of 5-ALA, the level of ROS was increased in a dose-dependent manner. Conclusion 5-ALA-PDT can affect the phagocytosis and the level of ROS of murine macrophage RAW264.7 cells, which may affect the efficacy of 5-ALA-PDT.
