摘  要:本研究主要目的是评价猪卵母细胞成熟培养中添加无机磷酸盐焦磷酸(PPi)对卵母细胞成熟、透明带泛素化水平及精卵结合能力的影响。试验设置对照组及0.1、1、5μg/m L 3个添加PPi处理组,检测卵母细胞成熟率、透明带泛素化水平及精卵结合情况。结果显示:1μg/m L PPi处理组卵母细胞成熟率为75.06%,显著高于对照组(P〈0.05);1μg/m L PPi处理组透明带泛素化水平显著高于其他各组(P〈0.05);1μg/m L PPi处理组透明带酶解时间为278.93 s,显著低于对照组(P〈0.05);1μg/m L PPi处理组精子黏附率为83.24,显著低于对照组(P〈0.05)。结论:PPi显著提高卵母细胞体外成熟率和透明带泛素化水平,降低透明带溶解时间和精子黏附率。This study was to evaluate the effect of inorganic phosphate pyrophosphate( PPi) addition on oocyte maturation,zona pellucida ubiquitination and sperm-oocyte binding ability during porcine oocyte maturation. The subjects were divided into four treatment groups of 0,0. 1,1,5 and 3μg/m L PPi supplementation in order to detect the oocyte maturation rate,the zona pellucida ubiquitination level,enzyme dissolving in zona pellucida and the sperm-oocyte binding. The results were that the oocyte maturation rate was 75. 06% in the 1 μg/m L PPi treatment group,significantly higher than the control group( P〈0. 05); the zona ubiquitination level in the 1 μg/m L PPi treatment group was significantly higher than in the other groups( P〈0. 05); the time for enzyme dissolving in zona pellucida was 278. 93 s in the 1 μg/m L PPi treatment group,significantly lower than in the control group( P〈0. 05); and the sperm-oocyte binding rate was 83. 24 in the 1 μg/m L PPi treatment group,significantly lower than in the control group( P〈0. 05). In conclusion,PPi supplementation significantly raised the rate of in vitro oocyte maturation and the level of zona pellucida ubiquitination,but reduced the time for dnzyme dissolving in zona pellucida and sperm-oocyte binding ability.
