摘  要:为掌握新疆南疆地区规模奶牛场牛病毒性腹泻病毒(BVDV)感染的病毒基因类型,更好地采取防控措施,本研究以BVDV基因高度保守的5'-UTR核苷酸序列设计并合成通用引物,提取疑似感染BVDV样品的核酸,采用一步法RT-PCR,扩增获得目的基因片段,经测序、比对、核苷酸同源性比较及构建基因遗传进化树。结果显示,30份疑似样品中,有2份样品PCR扩增阳性;测序分析显示,所检的阳性样品均为BVDV Ic型。研究表明,所调查的新疆南疆规模化奶牛场存在BVDV Ic型的感染,证实新疆地区牛场BVDV基因型存在多样性,为新疆规模奶牛场BVD病原鉴定、防控提供了理论依据。This study was to identify the genotype of bovine viral diarrhea virus on the large-scale cattle farms in southern Xinjiang. Specific primers were designed according to the published sequence of bovine viral diarrhea virus 5'-UTR gene. RNA samples were extracted and detected by RT-PCR with the special primers. The amplified fragments were sequenced. Analysis of the nucleotide sequences and construction of a phylogenetic tree were based on the 5'-UTR gene of bovine viral diarrhea virus with a biological software. The sequencing results showed that all the positive samples tested were of the BVDV Ic genotype,indicating BVDV Ic infection on large-scale cattle farms in southern Xinjiang,and that there were diverse genotypes of BVDVs there. The findings here provide theoretical basis for prevention and control of BVD pathogens on large-scale cattle farms in southern Xinjiang.
