摘　　要：目的:探讨弹性蛋白肽(EP)对髓样树突状细胞(mDCs)表面共刺激分子表达及炎症因子分泌的影响。方法:诱导小鼠骨髓来源的单个核细胞增殖分化为未成熟mDCs,随机分为空白对照组和EP组,空白对照组不加任何刺激,EP组加入终浓度为1mg/L EP。分别采用流式细胞术和qPCR技术检测mDCs表面共刺激分子CD80和CD86的表达,酶联免疫吸附试验(ELISA)法检测细胞上清液中白介素(IL)-6和转化生长因子(TGF)-β1的浓度。结果:流式细胞仪检测EP组CD80和CD86阳性表达率分别为(53.05±2.49)%、(50.05±3.90)%,显著高于空白对照组的(22.38±5.21)%、(19.33±5.80)%(P<0.05)。EP组CD80和CD86mRNA相对表达量及细胞上清液中IL-6和TGF-β水平均较空白对照组明显升高(P<0.05)。结论:EP可促进小鼠mDCs的活化及相关促炎因子的释放。Objective：To observe the differences in expression of cell surface co-stimulatory molecules and secretion of inflammatory factors by bone marrow-derived dendritic cells（DCs）in response to elastin peptide（EP）.Methods：We cultured bone marrow-derived DCs from BALB/c mice and infected with EP.The activation of myeloid DCs（mDCs）was characterized by studying expression of cell surface co-stimulatory molecules（CD80 and CD86）,release of transforming growth factor（TGF）-β1 and interleukin（IL）-12.Results：Flow cytometry analysis and qPCR detection showed significant up-regulation of CD80 and CD86 after treatment with EP（P〈0.05）.Meanwhile,ELISA revealed that the IL-6 and TGF-β1 levels in cell supernatant was conspicuous elevated（P〈0.05）.Conclusion：EP could promote the activation of mDCs in mice and the release of related pro-inflammatory factors.
