摘 要:目的:建立同时测定壮药华陀方中绿原酸、异嗪皮啶、迷迭香酸含量的方法。方法:采用HPLC法,色谱柱为依利特Sino Chrom ODS-BP柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.2%磷酸水溶液,采用梯度洗脱程序;流速为1.0 m L·min^-1;柱温:35℃;进样量10μL;检测波长为342 nm。结果:绿原酸、异嗪皮啶、迷迭香酸分别在61.00~610.0μg·m L^-1、5.000~50.00μg·m L^-1、5.000~50.00μg·m L^-1范围内与峰面积呈良好的线性关系,重复性好,平均回收率分别为98.92%(RSD=1.143%)、99.08%(RSD=1.717%)、99.88%(RSD=1.733%)。结论:所建立的方法操作简便,具有较好的精密度和重现性,可作为壮药华陀方的质量控制方法。Objective: To establish a method for the determination of chlorogenic acid,isofraxidin and rosmarinic acid in Zhuang drugs of Huatuo Formula. Methods: The HPLC was performed on the Sino Chrom ODS-BP column( 4. 6 mm × 250 mm,5μm) with the mobile phase of acetonitrile and 0. 2% phosphoric acid aqueous solution in gradient elution at a flow speed was 1. 0 m L·min^-1. The column temperature was 35 ℃. The injection volume was 10 μL,and the detection wavelength 342 nm. Results:The chlorogenic acid during the range of 61. 00-610. 0 μg·m L^-1 showed a good linear relation with the peak area( r = 0. 9999)and the average recovery rate was 98. 92%,with RSD of 1. 143%( n = 6). The isofraxidin during the range of 5. 000-50. 00 μg·m L^-1 showed a good linear relation with the peak area( r = 0. 9998) and the average recovery rate was 99. 08%,with RSD of1. 717%( n = 6). The rosmarinic acid during the range of 5. 000-50. 00 μg·m L^-1 showed a good linear relation with the peak area( r = 0. 9998) and the average recovery rate was 99. 88%,with RSD of 1. 733%( n = 6). Conclusion: The method is simple and fixable. It can be used for the quantity control of Zhuang drugs of Huatuo Formula.
