摘 要:目的:建立基于FT-NIR的连花清瘟胶囊中连翘苷的含量快速测定方法。方法:采集所有批次样品的FTNIR光谱,用多元散射校正法进行光谱预处理,采用偏最小二乘法(PLS)建立校正集模型,使用校正集内部交叉验证决定系数(R2)、验证集标准偏差与预测集标准偏差的比值(RPD)和校正集内部交叉验证均方根误差(RMSECV)作为评价指标比较不同方法处理后建模的准确性。采用验证集样品进行模型准确性再验证,以预测集均方根误差(RMSEP)作为评价模型的指标。利用建立的模型进行样品测定,测定结果和基于HPLC法测定结果进行比较。结果:不同预处理方式后建模所得到的R2、RPD和RMSECV表明以多元散射校正法处理效果最好;校正集样品均匀地分布在回归线的两侧,经内部交叉验证得RMSECV=0.351,RPD=3.94,R2=90.21;模型验证集预测均方根误差(RMSEP)为0.24。结论:基于FT-NIR所得结果与HPLC测定结果相比较,偏差较小。该方法具有简便、快速、准确、无损等特点,可用于连花清瘟胶囊中连翘苷含量的快速测定。Objective: It is aimed to establish a new analytical method for rapid determination of forsythin in Lianhua Qingwen Capsule based on FT-NIR and its application. Methods: We determined forsythin in all samples by high performance liquid chromatography. FT-NIR spectrums of all batches samples were collected and multiple scattering correction( MSC) was used to do the pretreatment of all spectrums. The partial least squares( PLS) was used to build a model. The R2,the ratio of the standard deviation between the validation sets and prediction sets( RPD),root-mean-squares error of cross-validation( RMSECV) were used to evaluate the accuracy of different approaches. The validation set samples to verify the accuracy of the model again,with root-mean-squares error of validation( RMSEP) as indicators of the model. Results: The effects of MSC process was the best.Calibration samples were evenly distributed on either side of the regression line,obtained by RMSECV = 0. 351,RPD = 3. 94,R2= 90. 21. The RMSEP was 0. 24. There were minimal differences between two different analytical methods. Conclusion: The method is simple,rapid,accurate,and can be used for rapid determination of forsythin content in Lianhua Qingwen Capsule.
