摘 要:目的探讨硫化氢(H2S)对大鼠急性百草枯(PQ)中毒肺过氧化损伤的影响。方法 30只SD大鼠分成对照组、模型组、治疗组。模型组、治疗组采用PQ(80mg/kg)灌胃染毒,模型组染毒后腹腔注射生理盐水,治疗组染毒后,腹腔注射NaHS(1.4μmol/kg),2次/d;对照组采用生理盐水灌胃,灌胃后腹腔注射生理盐水。72h后,观察肺组织病理改变;计算肺湿/干质量比值(W/D);测定肺组织匀浆内丙二醛(MDA)水平、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力。结果模型组肺组织损伤明显,治疗组较模型组减轻;模型组肺W/D较对照组增高(P<0.05),治疗组较模型组降低(P<0.05);模型组MDA高于对照组(P<0.05),SOD、GSH-Px低于对照组(P<0.05);治疗组中MDA较模型组降低(P<0.05),SOD、GSH-Px较模型组升高(P<0.05)。结论 H2S对急性PQ中毒大鼠肺过氧化损伤具有一定的保护作用。Objective To investigate the effects of hydrogen sulfide(H2 S)on the lung peroxidative damage in rats with acute paraquat(PQ)poisoning.Methods Thirty SD rats were divided into the control group,model group and treatment group.The model group and treatment group adopted PQ(80 mg/kg)gavage for poisoning contamination.The model group were intraperitoneally injected with normal saline after contamination.The treatment group was intraperitoneally injected by NaHS(1.4μmol/kg),two times a day.The control group adopted the normal saline gavage and intraperitoneally injected by normal saline after gavage.After 72 h,the pathological changes of lung tissue were observed.The lung wet/dry weight ratio(W/D)was calculated.The malondialdehyde(MDA)level in lung homogenate was detected,and the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were measured.Results The lung tissue injury in the model group was obvious,the treatment group was slighter than the model group;The lung W/D in the model group was significantly increased compared with the control group(P〈0.05),and the treatment group was decreased compared with the model group(P〈0.05);the MDA level in the model group was higher than that in the control group(P〈0.05),while the levels of SOD and GSH-Px were lower than those in the control group(P〈0.05);the MDA level in the treatment group was decreased compared with the model group(P〈0.05),while the levels of SOD and GSH-Px were increased compared with the model group(P〈0.05).Conclusion H2 S has a certain protective effect on the lung peroxidative damage in rats with acute PQ poisoning.
