摘 要:研究表明miR-17-92簇在诸多生命进程中发挥着重要作用。本研究目的是分析感染日本血吸虫小鼠肝脏不同时期miR-17-92簇的表达水平及其靶基因蛋白水平的表达变化,为进一步研究肝脏病理损害的分子机理奠定基础。q RT-PCR分析表明miR-17-92簇在感染日本血吸虫小鼠肝脏的不同感染时期呈现差异变化,随着时间延长呈现升高趋势;q RT-PCR和免疫印迹分析表明miR-17-92簇靶基因在感染日本血吸虫小鼠的肝脏呈现差异变化,进一步研究表明感染日本血吸虫小鼠血浆中miR-17-92簇的丰度也呈现差异变化。研究提示miR-17-92簇在血吸虫感染的终末宿主的肝脏病理损害中可能发挥重要的调控作用,也有可能成为血吸虫病引起宿主肝脏病变损伤诊断的新型标识分子。Recent studies indicated that miR-17-92 cluster plays an important role in many biological processes or diseases, such as cell development, immune response, cardiovascular diseases and neurodegenerative diseases. In the present study, we aim to determine the expression of miR-17-92 cluster and their targets in liver from mice infected with $chistosomajaponicum at different stages, qRT- PCR analysis indicated that the expression of miR-17-92 cluster in liver of mice infected with Schistosoma japonicum was increased in the later stage. Further analysis also indicated that the targets of miR-17-92 cluster in liver of mice infected with Schistosoma japonicum were altered. In addition, the abundance ofmiR-17-92 cluster in plasma of mice infected with Schistosomajaponicum also was changed, Overall, our preliminary results suggest that miR-17-92 cluster may be involved in pathological reactions of liver in a final host during schistosome infection. Moreover, our study also imply that miR-17-92 cluster may be a potential biomarker for indicating the progress of hepatic schistosomiasis.
